淫羊藿提取物HPLC特征图谱的构建及含量测定 被引量:1

Establishment of HPLC Characteristic Spectrum of Epimedium Extract and Quantitative Analysis on Its Main Chemical Constituents

摘要 为全面质量评价饲料添加剂淫羊藿提取物,用高效液相色谱(HPLC)方法构建不同品种及地区淫羊藿提取物的特征图谱,并测定其中5个质量控制成分的含量。以1 mL/min的流速,乙腈-水的流动相,35℃的柱温,270 nm的检测波长,20μL的进样量为条件,用Waters C18(4.6 mm×250 mm,5μm)的柱子上机。建立18批淫羊藿提取物的特征图谱,用外标法测定其中5个成分含量。建立的淫羊藿提取物特征图谱定有7个共有特征峰,基于高分辨数据分析和标准品比对,指认5个峰。采用聚类分析及主成分分析(PCA),分析了不同淫羊藿提取物的关联性。测定10批市场销售的淫羊藿提取物产品中五种黄酮醇苷的含量。建立的淫羊藿提取物特征图谱及含量测定方法符合方法学要求,为市售淫羊藿提取物的质量控制管理和资源开发提供了理论依据。 To comprehensively evaluate the quality of epimedium extract as feed additive,HPLC method was used to construct the characteristic spectrum of epimedium extract with various sources,and a quan⁃titative analysis method also development for its main chemical constituents based on HPLC.A waters C18 column was used with mobile phase consisted of acetonitrile-water at the flow rate of 1 mL/min.The column temperature was 35℃,the detection wavelength was set at 270 nm and the injection volume was 20μL.The HPLC characteristic spectrum was established based on 18 batches of epimedium ex⁃tracts with various sources,including different species and place of origin.Meanwhile,the contents of 5 main chemical constituents was determined by external standard method.The characteristic spec⁃trum of epimedium extracts were successfully es⁃tablished.A total of seven characteristic peaks were picked and five chromatographic peaks were identified by confirming with high resolution mass spectrum and reference substances.The correla⁃tion of the quality of epimedium extracts from dif⁃ferent sources was defined by cluster analysis and principal component.The content determination of five flavonol glycosides were determined based on 10 batches of epimedium extract products in the market.The established characteristic spectrum of epimedi⁃um extracts and content determination method could-meet the of methodological requirements,and pro⁃vide reference for the resource development and quality control of Epimedium extract in the market.
