脐血间充质干细胞移植对脑梗塞大鼠模型神经功能的影响

摘要 目的 探讨人脐血间充质干细胞移植对脑梗死大鼠神经功能的影响。方法 选取48只SD大鼠,随机分为4组,每组12只,分别为空白组、模型组、PBS组和移植组。空白组为正常老鼠,不做任何处理,其余3组均采用线栓法制作脑梗死大鼠模型,模型组只做成大鼠大脑中动脉栓塞(middle cerebral artery occlusion, MCAO)模型,移植组于造模成功后1 d经鞘内移植含1×106个脐血间充质干细胞的细胞悬液10μL;PBS组经同样方法注射10μL PBS溶液。移植后第1、3、7天进行神经功能评分,于第3、7天进行尼氏体计数、TUNEL法检测神经元细胞凋亡率、TTC评价梗死体积。结果 (1)与空白组相比,模型组和PBS组的神经功能评分、梗死体积及细胞凋亡数明显增加,尼氏体明显减少(P<0.05);(2)与模型组和PBS组相比,移植组梗死体积明显减小,细胞凋亡减少,尼氏体明显增多,神经功能评分明显改善(P<0.05)。结论 脐血间充质干细胞通过抑制细胞凋亡,缩小梗死体积,有效促进了脑梗死大鼠神经功能的恢复。 Objective To investigate the effect of human umbilical cord blood mesenchymal stem cell transplantation on nerve function in rats with cerebral infarction.Methods 48 SD rats were randomly divided into 4 groups with 12 rats in each group,which were blank group,model group,PBS group and transplantation group,respectively.The blank group was normal mice without any treatment,and the other three groups were made into cerebral infarction rat models by thread embolism.The model group was only made into MCAO model,and the transplantation group was transscribed 10μL of cell suspension containing 1×106 umbilical cord blood mesenchymal stem cells by intrathecal transplantation 1 day after the successful modeling;PBS group was injected with 10μL PBS solution by the same method.Neurological function scores were performed on the 1st,3rd and 7th day after transplantation,and neuronal cell apoptosis rate and infarct volume were evaluated on the 3rd and 7th day by Nignite count,TUNEL method and TTC method.Results Compared with the blank group,the neurological function score,infarct volume and apoptosis number of the model group and the control group were significantly increased,and Nissl bodies were significantly reduced(P<0.05);compared with the model group and the control group,in the transplantation group,the infarct volume was significantly reduced,cell apoptosis was reduced,Nissl bodies were significantly increased,and the neurological function score was significantly improved(P<0.05).Conclusion Cord blood mesenchymal stem cells can effectively inhibit cell apoptosis and reduce infarct volume,and promote the recovery of nerve function in rats with cerebral infarction.
