肌肽对2型糖尿病大鼠肾脏Hippo信号通路的影响 被引量:1

摘要 目的 通过以腹腔注射链尿佐菌素(streptozotocin, STZ)建立2型糖尿病肾病大鼠模型,探究肌肽通过抗氧化及改善Hippo通路对2型糖尿病大鼠肾脏的保护作用。方法 将60只SD雄性大鼠随机分成对照组、模型组、低剂量肌肽保护组、中剂量肌肽保护组和高剂量肌肽保护组共5组,利用试剂盒检测超氧化物歧化酶的活性;HE染色观察大鼠肾脏形态学变化,TUNNEL染色观察大鼠肾脏细胞凋亡,Western Blot检测相关蛋白的表达。结果 与模型组相比,肌肽保护组的大鼠肾脏超氧化物歧化酶的活性明显升高(P<0.05);HE染色结果显示,肌肽保护组的肾组织显微结构得到改善;TUNNEL染色结果显示,肌肽保护组的细胞凋亡率明显降低(P<0.05);Western Blot结果显示,与模型组相比,肌肽保护组yes相关蛋白(yes associated protein, YAP)水平明显降低(P<0.05)。结论 肌肽对糖尿病大鼠肾脏具有一定的保护作用,这种保护作用可能与其抗氧化性以及能够改善Hippo信号通路有关。 Objective The rat model of type 2 diabetic nephropathy was established by intraperitoneal injection of STZ to explore the protective effect of carnosine on the kidney of type 2 diabetic rats through antioxidant and Hippo pathway improvement.Methods 60 male SD rats were randomly divided into five groups:control group,model group,low dose carnosine protection group,medium dose carnosine protection group and high dose carnosine protection group,and the activity of superoxide dismutase was detected by the kit;HE staining was used to observe the morphological changes of rat kidney,TUNNEL staining was used to observe the apoptosis of rat kidney cells,and western blot was used to detect the expression of related proteins.Results Compared with the model group,the activity of renal superoxide dismutase in the carnosine protection group was significantly higher(P<0.05);HE staining showed that the renal tissue microstructure of carnosine protection group was improved;TUNNEL staining showed that the apoptosis rate of carnosine protection group was significantly decreased(P<0.05);western blot showed that compared with the model group,the level of YAP in the carnosine protection group was significantly lower(P<0.05).Conclusion Carnosine has a certain protective effect on the kidney of diabetic rats,which may be related to its antioxidant activity and the improvement of Hippo signaling pathway.
