摘要 [目的]研究α-酮戊二酸(AKG)对猪精子获能过程中运动状态、糖代谢及抗氧化能力的影响,为体外获能试验模型提供理论和科学依据。[方法]将长白猪精子分为7组,分别为新鲜精子组(Fresh)、获能对照组(0 mmol/L AKG)及获能AKG处理组(5、15、30、45、60 mmol/L),检测精子活率、顶体完整率、质膜完整性、活力、运动速率、ATP含量及乳酸含量,筛选出最适AKG浓度,进一步测定最适浓度处理后抗氧化指标的变化。[结果]AKG对获能精子的活率、顶体完整率、质膜完整性、活力均无显著影响,但能显著提高精子的运动速率、ATP和乳酸含量,且AKG浓度为15 mmol/L时ATP和乳酸含量升高效果最佳。与获能对照组相比,15 mmol/L AKG获能组的总抗氧化能力、超氧化物歧化酶和过氧化氢酶活性显著增加,丙二醛含量显著降低。[结论]添加AKG能够提高获能猪精子的运动速率、糖代谢水平及抗氧化能力。 [Objectives]The effects of a-ketoglutarate(AKG)on the motility,glucose metabolism and antioxidant capacity of pig spermatozoa during capacitation were studied to provide a theoretical and scientific basis for the test model of energization in vitro.[Methods]The sperm from Landrace pigs were divided into 7 groups including fresh sperm group,capacitation control group(0 mmol/L AKG)and capacitation AKG-treated groups(5,15,30,45 and 60 mmol/L).The sperm viability,acrosome integrity,plasma membrane integrity,motility,movement parameters,the content of ATP and lactic acid were measured to screen out the optimal concentration of AKG,and further determine the changes in antioxidant indexes after treatment with the optimal concentration of AKG.[Results]AKG had no significant effects on viability,acrosome integrity,plasma membrane integrity and motility of capacitation sperm,but significantly increased the rate of motility,the content of ATP and lactic acid.The effect of increasing the content of ATP and lactate was the best when the concentration of AKG was 15 mmol/L.Compared to the capacitation control group,the total antioxidant capacity,superoxide dismutase activity,and catalase activity were significantly increased,while the content of malondialdehyde was significantly decreased in the 15 mmol/L AKG capacitation group.[Conclusions]Adding AKG can improve the rate of motility,the level of glucose metabolism,and antioxidant capacity of pig sperm during capacitation.
