艾管针对缺血性脑卒中大鼠影响机制研究 被引量:1

摘要 目的:探究艾管针对缺血性脑卒中大鼠影响的机制。方法:采用“针控线栓法”对大鼠进行缺血性脑卒中造模,造模成功后随机分为模型组、针刺组与艾管针组,模型组不予治疗,常规饲养,针刺组对百会穴及左右旁开4 mm的穴位进行针刺,艾管针组是在针刺的基础上结合选取两侧的风池穴和供血穴进行艾管针治疗,在治疗前和治疗后7 d、14 d对各组大鼠的神经功能和行为学功能进行统计分析;在治疗后14 d对FGF-2、NCAM mRNA的阳性表达结果进行统计分析;在治疗后的7 d、14 d对各组大鼠的脑组织Dil^(+)/Brdu^(+)/NeuN^(+)、Dil^(+)/Brdu^(+)/GFAP^(+)细胞计数进行比较分析。结果:治疗7 d后及14 d后,艾管针组在神经功能评分、行为学功能观察评分、激光共聚焦显微镜观察大鼠脑组织Dil^(+)/Brdu^(+)/NeuN^(+)、Dil^(+)/Brdu^(+)/GFAP^(+)细胞计数表达等方面优于模型组和针刺组(P<0.05或P<0.01);治疗14 d后,荧光定量PCR检测FGF-2、NCAM mRNA表达,艾管针组优于模型组和针刺组(P<0.05或P<0.01)。结论:艾管针疗法不仅能促进缺血性脑卒中大鼠神经、行为功能的恢复,还能促进神经干细胞的增殖分化,减少神经细胞的损伤。 Objective:To explore the mechanism of moxa tube acupuncture in treatment of ischemic stroke in rats.Methods:The rats were established the model of ischemic stroke by using‘needle-control thread embolization method’.After successful modeling,the rats were randomly divided into the model group,the acupuncture group,and the moxa tube acupuncture group.The model group was given no treatment and was fed routinely.The acupuncture group was treated with needling Baihui(DU20)and the points 4mm lateral to DU20 on both sides;on which basis,the moxa tube acupuncture group was also treated with moxa tube acupuncture on bilateral Fengchi(GB20)and Gongxue point.Statistical analysis was performed on neurological and behavioral functions of the rats in each group before the treatment,7 days and 14 days after the treatment.The positive mRNA expressions of FGF-2 and NCAM were detected by fluorescence quantitative PCR and statistically analyzed after 14 days of the treatment.The cell counts of Dil^(+)/Brdu^(+)/NeuN^(+)and Dil^(+)/Brdu^(+)/GFAP^(+)in the brain tissues of each group were observed by laser confocal microscope and compared among the groups after 7 days and 14 days of the treatment.Results:After 7 days of the treatment and 14 days of the treatment,the neurological function score,the behavioral function score,as well as the cell counts of Dil^(+)/Brdu^(+)/NeuN^(+)and Dil^(+)/Brdu^(+)/GFAP^(+)were better in the moxa tube acupuncture group than those in the model group and in the acupuncture group(P<0.05,P<0.01).After 14 days of the treatment,the mRNA expressions of FGF-2 and NCAM were significantly improved in the moxa tube acupuncture group than those in the model group and in the acupuncture group(P<0.05,P<0.01).Conclusion:Moxa tube acupuncture can not only promote the recovery of neurological and behavioral functions in the rats with ischemic stroke,but also promote the proliferation and the differentiation of neural stem cells and reduce the nerve cell damage.
