摘要
目的:利用本课题组专利方法提取灵芝粗多糖,研究灵芝粗多糖对急性酒精性肝损伤的保护作用。方法:将50只雄性ICR小鼠随机分为正常组、模型组、联苯双酯组(150 mg/kg)和灵芝粗多糖高、低剂量组(36 mg/kg、7.2 mg/kg),每组10只。连续灌胃7 d,第8天,通过灌胃56°北京红星二锅头来建立小鼠急性酒精性肝损伤模型。计算小鼠肝脏指数,测定血清及肝脏组织相关生化指标,采用苏木精-伊红(HE)染色法观察小鼠肝脏组织病理学改变。结果:按照专利方法提取出来的灵芝粗多糖含量显著高于单用高压提取出来的灵芝粗多糖含量。动物实验,各组肝脏指数无显著性差异(P>0.05)。与模型组比较,灵芝粗多糖低、高剂量组可显著降低小鼠血清AST、ALT酶活性(P<0.01)以及肝脏组织MDA含量(P<0.01),显著提高肝脏组织GSH-PX、SOD酶活性(P<0.01),并对肝脏组织病理学变化有较大改善。结论:按照专利方法从灵芝子实体中提取灵芝粗多糖,能够获得更高含量的灵芝粗多糖,并且使用专利方法提取出来的灵芝粗多糖,对小鼠急性酒精性肝损伤具有一定的保护作用,其机制可能是通过对抗氧化应激。
Objective:To study the protective effect of Ganoderma lucidum crude polysaccharide on acute alcoholic liver injury by using a patented method of our group to extract Ganoderma lucidum crude polysaccharide.Methods:Fifty male ICR mice were randomly divided into normal group,model group,biphenyldiphenhydramine group(150 mg/kg)and Ganoderma crude polysaccharide high and low dose groups(36 mg/kg,7.2 mg/kg),10 mice in each group.The acute alcoholic liver injury model in mice was established by gavage of 56°Beijing Hongxing Erguotou for 7 d and on the 8th day.The liver index of mice was calculated,the biochemical indexes related to serum and liver tissues were measured,and the histopathological changes of mice liver were observed by hematoxylin-eosin(HE)staining method.Results:The crude polysaccharide content of Ganoderma lucidum extracted according to the patented method was significantly higher than that of Ganoderma lucidum extracted by high pressure alone.In animal experiments,there was no significant difference in liver index between the groups(P>0.05).Compared with the model group,the low and high dose groups of Ganoderma lucidum crude polysaccharide significantly reduced serum AST and ALT enzyme activities(P<0.01)as well as liver tissue MDA content(P<0.01),significantly increased liver tissue GSH-PX and SOD enzyme activities(P<0.01),and showed greater improvement on liver histopathological changes.Conclusion:The extraction of Ganoderma lucidum crude polysaccharide from Ganoderma lucidum seeds according to the patented method was able to obtain a higher content of Ganoderma lucidum crude polysaccharide,and the crude polysaccharide from Ganoderma lucidum extracted using the patented method had a protective effect against acute alcoholic liver injury in mice,probably through the mechanism of counteracting oxidative stress.
