摘要
目的:探讨硝唑尼特对咪喹莫特诱导的小鼠银屑病样皮炎的干预作用及其机制。方法:30只健康雌性BALB/c小鼠随机分为空白对照组、模型组、硝唑尼特组(100 mg/kg)3组,每组10只,利用5%咪喹莫特乳膏涂抹小鼠背部制备银屑病样皮炎模型,模型制备成功后观察小鼠受损皮肤情况并进行PASI评分,检测小鼠脾指数变化情况,对小鼠皮肤进行HE染色观察皮损情况,并采用ELISA法测定小鼠皮肤内炎症因子分泌的变化。结果:与空白对照组比较,模型组小鼠PASI评分明显增加(P<0.001),脾指数明显增加,血清IL-17和IL-23水平明显升高(P<0.01),病理切片结果显示皮肤增厚;与模型组比较,硝唑尼特组小鼠银屑病样皮损减少,PASI评分明显减少(P<0.01),脾脏指数显著下降(P<0.01),小鼠血清IL-17和IL-23水平明显下调(P<0.01)。结论:硝唑尼特通过降低皮肤中IL-17和IL-23的分泌来改善小鼠银屑病样皮炎。
Objective:To investigate the intervention effect of Nitazoxanide on imiquimod-induced psoriasis-like dermatitis in mice and its mechanism.Methods:Thirty healthy female BALB/c mice were randomly divided into blank control group,model group and nitazoxanide group(100 mg/kg),10 mice in each group.After the model was prepared,the damaged skin of mice was observed and PASI score was performed.The changes of splenic index in mice were detected,HE staining of mouse skin was performed to observe the skin lesions,and the changes of inflammatory factor secretion in mouse skin were measured by ELISA.Results:Compared with the blank control group,mice in the model group showed a significant increase in PASI score(P<0.001),a significant increase in spleen index,a significant increase in serum IL-17 and IL-23 levels(P<0.01),and thickened skin on pathological sections;compared with the model group,mice in the nitazoxanide group showed a decrease in psoriasis-like skin lesions,a significant decrease in PASI score(P<0.01),a significant decrease in spleen index decreased significantly(P<0.01),and serum IL-17 and IL-23 levels were significantly lowered(P<0.01)in mice.Conclusions:Nitazoxanide ameliorated psoriasis-like dermatitis in mice by decreasing the secretion of IL-17 and IL-23 in the skin.
