黑顶黄堇化学成分及其Cdc25A/Cdc25B酶抑制活性

摘要 目的研究黑顶黄堇Corydalis nigro-apiculata C.Y.Wu化学成分及其Cdc25A/Cdc25B酶抑制活性。方法黑顶黄堇95%乙醇提取物采用硅胶柱、Sephadex LH-20、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用全自动多功能酶标仪测定不同极性部位提取物对组蛋白磷酸化酶Cdc25A/Cdc25B的抑制效果。结果从中分离得到16个化合物,分别鉴定为mucroniferanine A(1)、mucroniferanine C(2)、mucroniferanine D(3)、mucroniferanine F(4)、山缘草定碱(5)、山缘草碱(6)、二氢血根碱(7)、6-甲氧基二氢血根碱(8)、血根碱(9)、乙氧基血根碱(10)、黄连碱(11)、原阿片碱(12)、巴马亭(13)、紫堇碱(14)、hendersine B methyl ester(15)、tomentelline C(16)。黑顶黄堇氯仿部位、正丁醇部位和水相萃取部位对于Cdc25A酶和Cdc25B酶均存在较好的抑制效果,其中正丁醇和水部位最为显著。结论化合物1~16均为首次从该植物中分离得到,该药材氯仿部位及正丁醇部位对组蛋白磷酸化酶Cdc25A/Cdc25B具有抑制效果。 AIM To study the chemical constituents from Corydalis nigro-apiculata C.Y.Wu and their Cdc25A/Cdc25B enzyme inhibitory activities.METHODS The 95%ethanol extract from C.nigro-apiculata was isolated and purified by silica,Sephadex LH-20 and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The inhibitory effect of the extracts of different polar fractions on the histone phosphorylase Cdc25A/Cdc25B was determined by an automatic multifunctional enzyme labeling instrument.RESULTS Sixteen compounds were isolated and identified as mucroniferanine A(1),mucroniferanine C(2),mucroniferanine D(3),mucroniferanine F(4),adlumidine(5),adlumine(6),dihydrosanguinarine(7),6-methoxydihydrosanguinarine(8),sanguinarine(9),ethoxysanguinarine(10),coptisine(11),protopine(12),palmatine(13),corydaline(14),hendersine B methyl ester(15),tomentelline C(16).The chloroform,n-butanol and water extraction sites showed better inhibition effects on Cdc25A enzyme and Cdc25B enzyme,among which n-butanol and water sites were the most significant.CONCLUSION Compounds 1-16 are isolated from this plant for the first time.The chloroform composition and n-butanol composition of this medicinal material have inhibitory effect on histone phosphorylase Cdc25A/Cdc25B.
