摘要
目的建立新疆地区大籽蒿的质量标准。方法采用显微法、TLC法进行定性鉴别;根据2020年版《中国药典》相关方法检查其水分、总灰分、醇浸出物含量以及挥发油含量,同时采用盐析辅助法对挥发油的提取方法进行优化;HPLC法测定绿原酸含量。结果显微特征明显,TLC显示清晰斑点,分离度好。初步拟定大籽蒿常规检查项下的限量标准为水分不高于9.00%,总灰分不超过10.00%,稀乙醇浸出物不少于10.00%,挥发油含量不少于0.60%。HPLC法考察结果显示,绿原酸在范围内线性关系良好(r=0.9998),平均加样回收率101.98%,RSD 1.41%;拟定该药材绿原酸含量不少于0.40 mg/g。结论该方法合理可行,可用于新疆地区产大籽蒿药材的质量控制。
AIM To establish the quality standard of Artemisia sieversiana Ehrhart ex Willd.locally grown in Xinjiang.METHODS A.sieversiana had its qualitative identification performed by microscopic method and TLC;its contents of moisture,total ash,alcohol-soluble extract and volatile oil determined complying with method of Chinese Pharmacopoeia(2020 edition);its volatile oil extraction improved by using salting out-assisted steam distillation;and its content of chlorogenic acid determined by HPLC method.RESULTS We obtained obviously microscopic features,and clear and well separated TLC spots.The initial quantitative limits were proposed to control the quality of A.sieversiana:no more than 9.00%moisture,10.00%of total ash and 10.00%diluted ethanol extract;and no lower than 0.60%volatile oil.From the result of HPLC,chlorogenic acid showed good linear relationships within its own ranges(r=0.9998),with 101.98%average recovery and 1.41%RSD.Therefore,we suggest no less than 0.40 mg/g chlorogenic acid in the herb.CONCLUSION This reasonable and feasible method can be used for quality control of A.sieversiana locally grown in Xinjiang.
