西兰花提取液对衰老模型小鼠抗氧化作用的影响

摘要 目的探究西兰花提取液对D-半乳糖致衰老小鼠抗氧化作用及血脂水平的影响。方法将60只SPF级小鼠随机分为空白对照组、衰老模型组、阳性对照组及西兰花提取液低、中、高剂量组,每组10只,除空白对照组外,其他组采用D-半乳糖构建小鼠衰老氧化模型,造模的同时,阳性对照组给予维生素C(300 mg·kg^(-1)),衰老模型组和空白对照组给予生理盐水,西兰花低、中、高剂量组分别给予西兰花提取液2.5 g·kg^(-1)、5.0 g·kg^(-1)、10.0 g·kg^(-1)灌胃,给药6周后,用比色法测定血清、肝组织及脑组织超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)和丙二醛(MDA)含量,用生化法测定血清甘油三酯(TG)、总胆固醇(CHO)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)的含量。结果应用SPSS 19.0软件进行统计分析,多组间均数比较采用单因素方差分析。结果各组间血清SOD活性(F=8.862,P<0.001)和GSH(F=9.947,P<0.001)、MDA(F=6.623,P<0.001)含量差异均有统计学意义。各组间肝组织SOD活性(F=5.133,P<0.001)和GSH(F=2.676,P<0.05)、MDA(F=13.467,P<0.001)含量差异均有统计学意义。各组间脑组织SOD活性(F=3.709,P<0.01)和GSH(F=3.892,P<0.01)、MDA(F=2.896,P<0.05)含量差异均有统计学意义。与正常对照组相比,衰老模型组血清、肝组织与脑组织中的SOD活性、GSH含量降低,MDA含量升高,差异均有统计学意义(P<0.01);与模型组相比,西兰花中、高剂量组血清、肝组织、脑组织SOD活性、GSH含量升高,MDA含量降低,差异有统计学意义(P<0.05或0.01);西兰花高剂量组血清TG、LDL水平降低,差异有统计学意义(P<0.05或0.01)。结论西兰花提取液具有抗氧化、降血脂的作用,这对延缓衰老、预防心脑血管等疾病具有重要的生理意义。 Objective To explore the effect of broccoli extract on antioxidation and blood lipid level of aging mice induced by D-galactose.Methods 60 SPF mice were randomly divided into blank control group,aging model group,positive control group and low-dose broccoli group,medium-dose broccoli group and high-dose broccoli group,with 10 mice in each group.In addition to the blank control group,D-galactose was used in the other groups to construct the aging oxidation model of mice.At the same time,the positive control group were given vitamin C(300 mg·kg^(-1)),the aging model group and blank control group were given normal saline,and the low-dose broccoli group,the medium-dose broccoli group and the high-dose broccoli group were given broccoli extract 2.5 g·kg^(-1),5.0 g·kg^(-1),and 10.0 g·kg^(-1)by intragastric administration,respectively.After 6 weeks of administration,the activity of superoxide dismutase(SOD)as well as the contents of glutathione(GSH)and malondialdehyde(MDA)in serum,liver and brain tissues were determined by colorimetry.And the contents of serum triglyceride(TG),total cholesterol(CHO),low density lipoprotein(LDL)and high density lipoprotein(HDL)in serum were measured by biochemical method.The results were analyzed with SPSS 19.0 software,and mean between multiple groups was compared with one-way ANOVA.Results There were significant differences in the activity of SOD(F=8.862,P<0.001),the contents of GSH(F=9.947,P<0.001)and MDA(F=6.623,P<0.001)in serum among groups.The activity of SOD(F=5.133,P<0.001)and the contents of GSH(F=2.676,P<0.05)and MDA(F=13.467,P<0.001)in liver tissues were statistically significantly different among the groups.The activity of SOD(F=3.709,P<0.01)and the contents of GSH(F=3.892,P<0.01)and MDA(F=2.896,P<0.05)in brain tissues were significantly different among the groups.Compared with the normal control group,the SOD activity and GSH contents in serum,liver and brain tissues of aging model group decreased,while the MDA contents increased,and the differences were statistically significant(P<0.01).Compared with the model group,the SOD activity and GSH contents in serum,liver and brain tissues of the the medium-dose broccoli group and the high-dose broccoli group increased,while the MDA contents decreased,and the differences were statistically significant(P<0.05 or 0.01).The serum levels of TG and LDL in the high-dose broccoli group significantly decreased,and the differences were statistically significant(P<0.05 or 0.01).Conclusion Broccoli extract has the function of antioxidation and blood lipid lowering,which is of great physiological significance to delay aging and prevent cardiovascular and cerebrovascular diseases.
