扶脾益肾汤调节Wnt/β-catenin信号通路对牙周炎大鼠牙周组织损伤的影响 被引量:4

Influence of Fupi Yishen Decoction on periodontal tissue damage in rats with periodontitis by regulating Wnt/β-catenin signaling pathway

摘要 目的观察扶脾益肾汤通过调节Wnt/β-catenin信号通路减轻牙周炎大鼠牙周组织损伤的作用机制。方法将72只SPF级SD大鼠随机分为对照组、模型组及扶脾益肾汤低(10g/kg)、中(20g/kg)、高(40g/kg)组、扶脾益肾汤高(40 g/kg)+通路抑制剂Wnt-C59(30 mg/kg)组(高+Wnt-C59组)。每组12只,用细菌感染建立牙周炎大鼠模型。给药结束后,检测牙周指标菌斑指数(PLI)、牙周探诊深度(PD)、龈沟出血指数(SBI);Micro-CT检测牙槽骨吸收;HE染色观察牙周组织病理学变化;TRAP染色分析破骨细胞数目;Western blot检测Wnt/β-catenin通路蛋白表达。结果与对照组比较,模型组大鼠牙槽骨吸收、牙周结构破坏严重,PLI、PD、SBI、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)水平、破骨细胞数目表达显著升高(P<0.05),Wnt、β-catenin蛋白表达显著下降(P<0.05)。与模型组比较,扶脾益肾汤各处理组牙周结构改善,PLI、PD、SBI、TNF-α、IL-6、IL-1β水平、破骨细胞数目表达显著下降(P<0.05),Wnt、β-catenin蛋白表达显著上升(P<0.05)。结论扶脾益肾汤可能通过激活Wnt/β-catenin信号通路,减轻牙周炎大鼠牙周组织损伤。 Objective To observe the mechanism of Fupi Yishen Decoction in reducing periodontal tissue damage in rats with periodontitis by regulating the Wnt/β-catenin signaling pathway.Methods Seventy-two SPF SD rats were randomly divided into control group,model group,Fupi Yishen Decoction low(10 g/kg),middle(20 g/kg),high(40 g/kg)groups,and Fupi Yishen Decoction high(40 g/kg)+pathway inhibitor Wnt-C59(30 mg/kg)group(high+Wnt-C59 group).The periodontitis rat model was established by bacterial infection.After administration,the periodontal index plaque index(PLI),periodontal probing depth(PD)and gingival crevicular bleeding index(SBI)were detected;Micro-CT was applied to detect alveolar bone resorption;HE staining was applied to observe the pathological changes of periodontal tissue;TRAP staining was applied to analyze the number of osteoclasts;Western blot was applied to detect the expression of Wnt/β-catenin pathway proteins in periodontal tissues.Results Compared with control group,the alveolar bone resorption and periodontal structure destruction of the rats in model group were severe,the PLI,PD,SBI,levels of tumor necrosis factor-α(TNF-a),interluekin-6(IL-6),IL-1β,the number of osteoclasts was obviously increased(P<0.05),the expressions of Wnt andβ-catenin proteins were obviously decreased(P<0.05);compared with model group,the periodontal structure of each treatment group with Fupi Yishen Decoction improved,the PLI,PD,SBI,levels of TNF-α,IL-6,IL-1β,the number of osteoclasts were obviously decreased(P<0.05),the expressions of Wnt andβ-catenin proteins were obviously increased(P<0.05).Conclusion Fupi Yishen Decoction may reduce the damage of periodontal tissue in rats with periodontitis by activating the Wnt/β-catenin signaling pathway.
