钙黏蛋白家族基因CDH3通过PI3K/AKT通路影响宫颈癌肿瘤细胞的生长 被引量:2

摘要 目的探究钙黏蛋白(cadherin,CDH)家族基因CDH3在宫颈癌发展中的作用。方法从TCGA数据库获取宫颈癌患者数据,通过R软件分析在宫颈癌组织中钙黏蛋白家族基因的表达情况,筛选出表达上调的基因并与宫颈癌预后相关基因取交集,确定目的基因;收集10例宫颈癌患者手术切除的癌组织和癌旁组织,用qPCR检测样本内目的基因表达水平;通过CCK8和Transwell实验检测目的基因表达水平对宫颈癌细胞系(HeLa和C33A)的增殖、迁移和侵袭能力的影响;通过Western blot实验和KEGG通路分析研究目的基因对PI3K/Akt信号通路的影响。结果CDH3在宫颈癌患者组织中高表达,其表达水平与宫颈癌患者的预后生存相关。qPCR结果显示CDH3在宫颈癌癌组织中的表达水平显著高于癌旁组织。敲低CDH3可抑制HeLa和C33A细胞的增殖、迁移和侵袭。CDH3的敲低和过表达可分别造成宫颈癌细胞中p-PI3K、p-Akt蛋白水平降低和增加。结论CDH3与宫颈癌的发展有关,可能通过激活PI3K/Akt信号通路促进宫颈癌细胞的增殖、迁移和侵袭。 Objective To investigate the role of cadherin family gene CDH3 in the development of cervical cancer.Methods The data of cervical cancer patients were obtained from TCGA database,and the expression of cadherin family genes in cervical cancer was analyzed using the R software.The up-regulated genes were screened and intersected with the prognostic genes of cervical cancer to obtain the target genes.In addition,surgically removed cervical cancer samples and paracancer tissues were collected from 10 patients,and the expression of target genes was detected by qPCR.The effects of target gene expression levels on the proliferation,migration and invasion of cervical cancer cell lines(HeLa and C33A)were evaluated by CCK8 and Transwell assays.The influences of target genes on PI3K/Akt signaling pathway were measured by Western blot and KEGG analysis.Results CDH3 was highly expressed in cervical cancer tissues,and its level was correlated with the prognosis and survival of patients.The results of qPCR assay showed that the expression of CDH3 in cervical cancer was significantly higher than that in paracancer tissues.CDH3 knockdown inhibited the proliferation,migration and invasion of cervical cancer cells.Furthermore,CDH3 knockdown and overexpression respectively decreased and increased p-PI3K and p-Akt protein expression in cervical cancer cells.Conclusion CDH3 is involved in the development of cervical cancer,and may promote the proliferation,migration and invasion of cervical cancer cells by activating PI3K/Akt signaling pathway.
