信号增强型汞离子荧光蛋白探针的设计及表征

摘要 重金属离子在化工产业、汽车制造、电子行业等领域应用越来越广泛,其导致的环境污染也愈发严峻,汞离子污染成为其中典型代表。汞作为一种有毒重金属,会严重危害人类的身体健康。因此,水环境中汞离子的检测与治理至关重要。该研究通过在荧光蛋白EGFP发色团附近引入一个Hg^(2+)结合位点,成功构建了一种荧光信号增强型的Hg^(2+)全细胞生物传感器(EGFP T203C S205C),并利用荧光光谱技术对其灵敏性和选择性进行表征。研究结果表明该生物传感器能够响应微摩尔级别的Hg^(2+),并且对Hg^(2+)具有良好的选择性和较强的抗干扰能力,是一种简单、高效、成本低廉的Hg^(2+)检测方法。 Heavy metal ions are more and more widely used in chemical chemistry,automobile manufacturing,electronic industry and other fields.The environmental pollution problem caused by heavy metal ions is becoming more and more serious,among which mercury ion pollution has become a typical representative.Mercury,as a toxic heavy metal,can seriously harm our human health.Therefore,the detection and treatment of mercury ions in water environment is very important to protect our human health.In this study,a Hg^(2+)-enhanced whole-cell biosensor(EGFP T203C S205C)was successfully constructed by introducing an Hg^(2+)binding site near the fluorescent protein EGFP chromophor,and its sensitivity and selectivity were characterized by fluorescence spectroscopy.The results show that the biosensor can respond to micromole level of Hg^(2+),and has good selectivity and strong anti-interference ability to Hg^(2+),which is a simple,efficient,and low-cost detection method for Hg^(2+).
