摘要
目的探讨Stathmin在白细胞介素-1β(IL-1β)调节人真皮成纤维细胞迁移中的作用及机制。方法使用IL-1β(10 ng/mL)处理细胞24 h模拟体外早期炎症反应,利用小干扰RNA干预Stathmin和FAK表达,通过细胞划痕试验观察细胞迁移,Western blot检测FAK和Stathmin蛋白表达情况。结果与对照组相比,IL-1β处理组细胞迁移明显增加,Stathmin和FAK蛋白表达明显增加;当下调Stathmin或FAK表达后,细胞迁移明显受抑;并且,下调Stathmin表达后,FAK蛋白表达明显降低。结论IL-1β可能通过调节Stathmin/FAK通路促进炎症早期人真皮成纤维细胞迁移。
Objective To investigate the role and mechanism of interleukin-1β(IL-1β)in the regulation of human dermal fibroblast migration by Stathmin.Methods Cells were treated with IL-1β(10 ng/mL)for 24 h to mimic the early inflammatory response in vitro,and small interfering RNA was used to interfere with Stathmin and FAK expression.Cell migration was observed by cell scratch assay,and FAK and Stathmin protein expression was detected by Western blot.Results Compared with the control group,cell migration was significantly increased in the IL-1β-treatment group,and Stathmin and FAK protein expression were significantly increased.When Stathmin or FAK expression was down-regulated,cell migration was significantly inhibited;when Stathmin expression was down-regulated,FAK protein expression was significantly decreased.Conclusion IL-1βmay promote human dermal fibroblast migration in early inflammation by regulating the Stathmin/FAK pathway.
