TWEAK/Fn14在结节性红斑患者皮损组织中的表达

Expression of TWEAK/Fn14 in Skin Lesion of Patients with Erythema Nodosum

摘要 目的探讨TWEAK/Fn14信号通路在结节性红斑中的表达情况及意义。方法收集西安交通大学第二附属医院皮肤科2021年6月-2021年10月结节性红斑患者15例,取同期正常组织为对照,行石蜡组织包埋术及病理切片,提取组织蛋白和RNA,免疫组织化学染色、实时荧光定量PCR及蛋白印迹检测TWEAK和Fn14分子在结节性红斑皮损组织和正常组织中的表达,并进行差异比较和统计学分析。结果免疫组织化学检测结果显示,TWEAK/Fn14蛋白在正常组织中低表达,在患者皮损处显著高表达,尤其在脂肪小叶及血管周围,并伴有多种炎症细胞浸润、脂肪肉芽肿和纤维化;实时荧光定量PCR及蛋白印迹检测显示患者皮损组织中的TWEAK/Fn14信号表达显著高于正常组织对照组(P<0.05)。结论TWEAK/Fn14信号通路可能在结节性红斑发病机制中发挥重要作用,有望成为治疗结节性红斑的潜在靶点。 Objective To investigate the expression and significance of TWEAK/Fn14 signaling pathway in erythema nodosum.Methods A total of 15 patients with erythema nodosum treated from the Department of Dermatology of the Second Affiliated Hospital of Xi′an Jiaotong University from June 2021 to October 2021 were enrolled.The normal tissues of the same period were taken as control.Tissues were processed with pathological processing for paraffin embedding by slicing into sections.Tissue protein and RNA were extracted,and then the expression of both TWEAK and Fn14 in skin lesion of erythema nodosum and normal tissues were determined by immunohistochemical staining,real-time fluorescence quantitative PCR and Western blot.Statistical analysis was performed for the differences.Results Immunohistochemical staining showed that TWEAK/Fn14 protein was lowly expressed in normal tissues,and significantly highly increased in skin lesions,especially in fat lobules and vessels,accompanied by a variety of inflammatory cell infiltration,fat granulomas and fibrosis.Real-time fluorescence quantitative PCR and Western blot analysis demonstrated that the expression levels of TWEAK and Fn14 in the skin lesions of the patients was significantly higher than those of the control group(P<0.05).Conclusion The TWEAK/Fn14 signaling pathway may play an important role in the pathogenesis of erythema nodosum,and is a potential therapeutic target for patients with erythema nodosum.
