高效液相色谱法测定2-辛烯基琥珀酸基团的研究

摘要 为验证高效液相色谱法测定辛烯基琥珀酸淀粉钠中2-辛烯基琥珀酸基团含量的可行性,并探究更优的高效液相色谱检测方法,利用高效液相色谱仪,通过测定辛烯基琥珀酸淀粉钠中游离2-辛烯基琥珀酸和总2-辛烯基琥珀酸的含量获得2-辛烯基琥珀酸基团含量,并对样品的前处理条件进行优化。结果显示,利用高效液相色谱法测定的2-辛烯基琥珀酸基团的定量限为0.8394μg/mL,对样品提取条件进行优化,确定了游离2-辛烯基琥珀酸的振荡提取时间为18 h,提取总2-辛烯基琥珀酸的氢氧化钾浓度为0.1 mol/L,水解时间为1 h,2种物质在优化条件下的回收率、精密度均良好,且2-辛烯基琥珀酸基团的测定结果稳定。因此,利用高效液相色谱可有效测定2-辛烯基琥珀酸基团的含量。文章探究了高效液相色谱测定辛烯基琥珀酸淀粉钠中2-辛烯基琥珀酸基团的检测方法,以期为2-辛烯基琥珀酸基团的高效检测提供参考方向。 In order to verify the feasibility of high performance liquid chromatography for the determination of 2-octenyl succinate group in sodium starch octenyl succinate,and to explore the better high performance liquid chromatography(HPLC)method,in this study,the content of 2-octenyl succinate group was obtained by determination of free 2-octenyl succinate and total 2-octenyl succinate in sodium starch octenyl succinate using high performance liquid chromatography,and the pretreatment conditions of the sample were optimized.The results showed that the limit of quantification of 2-octenyl succinate group determined by HPLC was 0.8394μg/mL.The extraction conditions of samples were optimized,the oscillatory extraction time of free 2-octenyl succinate was 18 h,the potassium hydroxide concentration of the total 2-octenyl succinate was 0.1 mol/L,and the hydrolysis time was 1 h.The recovery and precision of the two substances were good under the optimized conditions,the results of 2-octenyl succinate group determination were stable.Therefore,the content of 2-octenyl succinate group can be determined effectively by HPLC.In this study,a method for the determination of 2-octenyl succinate group in sodium starch octenyl succinate by high performance liquid chromatography was investigated,in order to provide a reference direction for the efficient detection of 2-octenyl succinate group.
