摘要
目的基于生物信息学分析ABAT基因在肝癌中的表达及功能探讨。方法利用在线Oncomine和GEPIA数据库分析ABAT在肝癌与正常肝组织的表达,同时在肝癌细胞株采用qPCR及Western blot法检测ABAT的表达;从NCBI-GEO数据库(https://www.ncbi.nlm.nih.gov/gds/)下载研究号为GSE14520的芯片数据,提取其中214例肝癌组织的基因表达谱及相关临床数据,基于ABAT基因表达的中位值分为高低表达组,分析2组与其对应的临床信息相关性及预后情况;TCGA数据库数据进行验证ABAT与肝癌的预后,体外实验初步探讨ABAT对肝癌的影响。结果ABAT在肝癌组低表达且与AFP、TNM分期、转移相关,其总生存率较高表达组差(P<0.05)。体外实验:ABAT抑制肝癌细胞的增殖、凋亡、迁移及侵袭能力(P<0.05),调控Wnt/β-catenin信号通路(P<0.05)。结论ABAT在肝癌患者中呈低表达,参与肝癌的进展,影响细胞的生长增殖、周期、细胞凋亡过程,其机制可能与Wnt/β-catenin信号通路相关。
Objective To investigate the expression and function of ABAT in liver cancer based on bioinformatic.Methods The Oncomine and GEPIA data were used to analyze the expression of ABAT in liver cancer and normal liver tissues,and the expression of ABAT in liver cancer cell lines was detected by qPCR and Western blot.Download the chip data GSE14520 from the NCBI-GEO database and collected the gene expression profiles and related clinical data of 214 liver cancer tissues.The correlation and prognosis of ABAT expression data and its clinical information were analyzed and the association of ABAT with prognosis was validated by TCGA database.Finally,the function of ABAT on liver cancer were investigated in vitro experiments.Results The expression of ABAT was lower in liver cancer and associated with AFP,TNM stage,metastasis(P<0.05).The samples with low ABAT expression showed a poor prognosis(P<0.05).Functional assays demonstrated that ABAT inhibited liver cancer cell proliferation,migration and invasion and regulated Wnt/β-catenin signaling pathway(P<0.05).Conclusion The expression of ABAT is lower in liver cancer patients,and impacted on the progression of liver cancer cells which may be related to the process of regulating cell proliferation,cell cycle,apoptosis,and Wnt/β-catenin signaling pathway.
