摘要
目的:观察理筋手法经β-catenin/PPARγ信号通路对大白兔竖脊肌脂肪浸润的影响,分析其作用机制。方法:将18只新西兰大白兔随机分为空白组、模型组及治疗组,每组6只,2 d治疗1次,连续6周。竖脊肌采用苏木精-伊红(HE)染色观察病理学变化,酶联免疫吸附测定(ELISA)法检测大白兔血清中白细胞介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)及脂肪因子瘦素(Leptin)、脂联素(Adiponectin,ADPN)的水平,蛋白免疫印迹法(Western Blot)检测竖脊肌组织中β-连环蛋白(β-catenin)、过氧化物酶体增殖物激活受体γ(PPARγ)蛋白表达水平。结果:HE染色结果显示:与空白组相比,模型组肌纤维数量减少、排列紊乱,肌束间隔增大,肌束内及肌束间脂肪细胞簇集增多,体积增大,理筋手法治疗组肌束间隔小于模型组,脂肪细胞数量及体积也小于模型组。ELISA法检测结果:与空白组相比,模型组大白兔血清Leptin、TNF-α水平明显升高,差异有统计学意义(P<0.05);IL-1水平显著升高,差异有统计学意义(P<0.01),而ADPN水平明显降低,差异有统计学意义(P<0.01)。与模型组相比,治疗组血清TNF-α、IL-1及Leptin水平明显降低,差异有统计学意义(P<0.05),而ADPN水平显著升高,差异有统计学意义(P<0.05)。Western Blot检测结果:与空白组相比,模型组中β-catenin蛋白显著降低,差异有统计学意义(P<0.01);与模型组相比,治疗组显著升高,差异有统计学意义(P<0.001);与空白组相比,模型组中PPARγ蛋白明显升高,差异有统计学意义(P<0.01);与模型组相比,治疗组中PPARγ蛋白显著降低,差异有统计学意义(P<0.01)。结论:理筋手法改善竖脊肌脂肪浸润,可能与调控β-catenin/PPARγ信号通路、调节炎症因子及脂肪因子水平有关。
Objective:To observe the effect of tendon manipulation on theβ-catenin/PPARγsignaling pathway in the muscle tissue of erector spinal muscle fat infiltration in white rabbit,and to analyze its mechanism of action.Methods:18 New Zealand white rabbits were randomly divided into blank group,model group and treatment group,with 6 rabbits in each group,and the treatment was once every other day for 6 weeks.Hematoxylin-eosin(HE)staining of erector spinal muscles was used to observe pathological changes,the levels of interleukin-1(IL-1),tumor necrosis factor-α(TNF-α),leptin and adiponectin(ADPN)in serum of white rabbits were detected by Enzyme-linked immunosorbent assay(ELISA).Western Blot was used to detect the expression levels ofβ-catenin and peroxisome proliferator-activated receptorγ(PPARγ)proteins in erector spinal muscle tissue.Results:Compared with the blank group,the number of myofibers in the model group was reduced,the arrangement was disordered,the fascicular septum increased,the cluster of adipocytes in and between the fasciculations increased,and the volume increased,the fascicular spacing in the tendon manipulation treatment group was smaller than that in the model group,and the number and volume of adipocytes were also smaller than those in the model group.ELISA test results:compared with the blank group,the serum leptin and TNF-αlevels in the model group were significantly increased(P<0.05),IL-1 levels were significantly increased(P<0.01),while the ADPN levels were significantly reduced(P<0.01).Compared with the model group,the serum TNF-α,IL-1 and leptin levels in the treatment group were significantly lower(P<0.05),while the ADPN levels were significantly increased(P<0.05).Western Blot detection results:compared with the blank group,theβ-catenin protein in the model group was significantly reduced(P<0.01),and compared with the model group,theβ-catenin protein in the treatment group significantly increased(P<0.001).Compared with the blank group,the PPARγprotein in the model group was significantly increased(P<0.01),and compared with the model group,the PPARγprotein in the treatment group was significantly reduced(P<0.01).Conclusion:The improvement of fat infiltration in erector spinal muscles by tendon manipulation may be related to the regulation ofβ-catenin/PPARγsignaling pathway and the regulation of inflammatory factors and adipose factor levels.
