姜黄素对小鼠溃疡性结肠炎疗效及免疫作用机制

Therapeutic effect and immune mechanism on ulcerative colitis treated by curcumin in mice

目的研究姜黄素(Cur)对葡聚糖硫酸钠(DSS)诱导小鼠急性溃疡性结肠炎(UC)的疗效及免疫作用机制。方法采用DSS灌胃造模,随机分为正常组(N组)、模型组(M组)、柳氮磺胺吡啶组(SASP组)和Cur高、中、低剂量组(Cur-H、Cur-M、Cur-L组),给予相应剂量药物,通过进行疾病活动指数(DAI)评分、结肠组织学观察、小鼠血清白细胞介素(IL)-17A、IL-2、IL-23、肿瘤坏死因子(TNF)-α的含量检测、碳粒廓清及结肠组织髓过氧化物酶(MPO)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)的水平测定来研究Cur对UC小鼠免疫作用机制的影响。结果经Cur治疗后,DAI评分降低、溃疡程度减轻、IL-2表达水平增加,IL-17A、IL-23、TNF-α表达水平降低,差异有统计学意义(P<0.05)。Cur-H组、Cur-M组脾脏指数、廓清指数k及校正吞噬指数α与M组差异有统计学意义(P<0.05,P<0.01,P<0.001)。结论对UC小鼠具有治疗作用的Cur可增强小鼠的免疫功能。

Objective To investigate the therapeutic effect and immune mechanism of curcumin(Cur)on acute ulcerative colitis(UC)induced by dextran sodium sulfate(DSS)in mice.Methods The model was established by DSS gavage and randomly divided into normal group(N group),model group(M group),sulfasalazine group(SASP group)and Cur high,medium and low dose(Cur-H,Cur-M,Cur-L)groups,each group was given corresponding doses of drugs.The influence of immune mechanism of Cur was evaluated on UC in mice by disease activity index(DAI)score,colonic histological observation,content detection in the level of interleukin(IL)-17A,IL-2,IL-23,tumor necrosis factor(TNF)-αof the mouse serum,carbon particle clearance and level determination of myeloperoxidase(MPO),malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)in colonic tissue.Results After Cur treatment,DAI score and the degree of ulceration decreased,the expression of IL-2 level increased,the expression levels of IL-17A,IL-23 and TNF-αdecreased,the difference was statistically significant(P<0.05).Compared with group M,there were significant differences in spleen index,clearance index k and corrected phagocytic indexαin Cur-H group and Cur-M group(P<0.05,P<0.01,P<0.001).Conclusions Cur has therapeutic effect on UC mice and could enhance the immune function of mice.
