摘要
目的:比较水蒸气蒸馏法(HD)和超声辅助提取法(UAE)提取香附挥发油的化学成分和抗氧化活性。方法:采用气相色谱-质谱联用(GC-MS)分析采用HD和UAE提取所得香附挥发油的化学成分,同时采用清除DPPH、ABTS+和羟基自由基的活性评价其抗氧化活性。结果:GC-MS分析显示,两种方法提取的香附挥发油主要成分是氧化倍半萜和倍半萜。HD所得挥发油的化学成分鉴定率为(92.17±3.21)%,UAE得到挥发油的化学成分鉴定率为(61.34±5.41)%。UAE和HD所得挥发油的DPPH、ABTS+和羟基自由基的IC50值分别为(23.82±4.51)、(36.01±1.86)、(49.07±5.51)μg/mL和(30.69±3.46)、(56.28±4.17)、(82.81±4.87)μg/mL。结论:HD和UAE提取所得香附挥发油的化学成分有一定的差异,UAE所得挥发油的抗氧化活性强于HD所得挥发油,可能与UAE提取所得挥发油中含有一些HD不包括且具有抗氧化活性的化学成分有关。
Objective:To compare the chemical composition and antioxidant activity of volatile oil from Cyperi Rhizoma by hydrodistillation(HD)and ultrasonic-assisted extraction(UAE).Methods:The volatile oil from Cyperi Rhizoma was extracted by HD and UAE,respectively.Then the components of the volatile oil extracted by the two methods were analyzed by gas chromatography-mass spectrometry(GC-MS)and the antioxidant activity of the volatile oil was assessed by DPPH,ABTS+and hydroxyl radical scavenging activity.Results:According to the GC-MS analysis,the major ingredients in the volatile oil extracted by HD and UAE were oxygenated sesquiterpenes and sesquiterpenes.(92.17+3.21)%of the chemical components were identified from the the volatile oil obtained by HD,while(61.34+5.41)%of the chemical components were identified from the the volatile oil obtained by UAE.The DPPH,ABTS*and hydroxyl radical scavenging ICso value of volatile oil obtained by UAE and HD were(23.82±4.51),(36.01±1.86),(49.07±5.51)μg/mL and(30.69±3.46),(56.28±4.17),(82.81±4.87)μg/mL,respectively.Conclusion:The chemical compositions of the volatile oil from Cyperi Rhizoma obtained by HD and UAE are obviously different.The antioxidant activity of volatile oil by UAE is stronger than that by HD.This might be the result of the substances obtained by UAE also contain some volatile components which have antioxidant activity.
