刺老苞不同炮制品对成肌细胞C2C12增殖活性和成分的影响

Effects of different processed products of Aralia echinocaulis on proliferation activity and components of myoblasts C2C12

目的:基于刺老苞不同炮制品对成肌细胞C2C12增殖活性的影响,利用UPLC-ESI-MS/MS方法探究不同炮制品存在药效差异的物质基础。方法:MTT法检测不同浓度(25、50、100、200μg/mL)刺老苞生品、清炒品、盐炙品对成肌细胞C2C12增殖的影响。采用UPLC-ESI-MS/MS方法分析3种刺老苞炮制品的化学成分,并对其进行主成分分析、正交偏最小二乘法判别分析、聚类分析等多元统计分析,初步推断不同刺老苞炮制品的成分差异。结果:MTT检测结果显示,在一定浓度范围内3种刺老苞炮制品均促进成肌细胞C2C12增殖,除了50μg/mL刺老苞生品和清炒品外,其余各浓度刺老苞生品、清炒品、盐炙品两两比较均具有显著性差异(P<0.01,P<0.05)。多元统计分析显示,3种炮制品存在药效差异可能与酚酸类、萜类、生物碱、黄酮类等成分有关。结论:该研究从药物的药效差异出发,结合代谢组学,初步阐述了刺老苞不同炮制品存在药效差异的物质基础,为民族药的进一步开发和应用奠定理论基础。

Objective:Based on the effect of different processed products of Aralia echinocaulis on the proliferation activity of myoblasts C2C12,the material basis of pharmacodynamic differences in different processed products was analyzed by UPLC-ESI-MS/MS.Methods:MTT assay was used to detect the effects of different concentrations(25,50,100,200μg/mL)of raw,fried and salt products on the proliferation of C2C12 cells.The chemical composition of different processed products of Aralia echinocaulis was analyzed by UPLC-ESI-MS/MS.Meanwhile,multiple statistical analysis methods including principal component analysis(PCA),orthogonal partial least squares discriminant analysis(OPLS-DA)and cluster analysis were applied to preliminarily infer the composition differences of different processed products of Aralia echinocaulis.Results:The results of MTT assay showed that three processed products of Aralia echinocaulis promoted the proliferation of myoblasts C2C12 in a certain concentration range.Except for the raw and fried products of Aralia echinocaulis at a concentration of 50μg/mL,there were significant differences in pairwise comparisons of different processed products at each concentration(P<0.01,P<0.05).Multivariate statistical analysis showed that there were pharmacodynamic differences among the three processed products that may be related to phenolic acids,terpenoids,alkaloids,flavonoids and other components.Conclusion:Based on the difference of drug efficacy and metabolomics,the material basis of the difference of drug efficacy in different processed products of Aralia echinocaulis is preliminarily expounded in this study,which lays a theoretical foundation for the further development and application of ethnic medicine.
