南、北五味子对离体大鼠气管平滑肌张力的影响 被引量:1

Effects of Northern and Southern Schisandra chinensis on Smooth Muscle Tension of Isolated Tracheal in Rat

摘要 目的探讨南、北五味子木脂素和多糖对离体大鼠气管平滑肌张力的影响.方法分离制备南、北五味子木脂素及多糖,采用高效液相色谱法(HPLC)和苯酚硫酸法测定木脂素及多糖含量;应用离体组织灌流实验方法分析南、北五味子木脂素和多糖对大鼠气管环静息状态、乙酰胆碱(ACh)和氯化钾(KCl)预收缩气管环张力的影响.结果南、北五味子中五味子酯甲、五味子甲素、五味子醇甲、五味子乙素、五味子醇乙、五味子丙素6种木脂素总含量分别为(18.63±0.89)mg/g和(19.33±0.95)mg/g,粗多糖含量分别为19.27 mg/g和20.5 mg/g;南、北五味子木脂素(0.25~1.75 mg/mL)和多糖(0.06~2.00 mg/mL)对静息状态下离体大鼠气管环的张力均未见明显影响;南、北五味子多糖(0.06~2.00 mg/mL)对ACh预收缩的气管环张力亦未见明显影响,而南、北五味子木脂素(0.25~1.75 mg/mL)可剂量依赖性抑制ACh和KCl诱导的气管环收缩;对ACh诱导收缩的最大抑制率分别为(68.21±10.08)%和(71.72±9.80)%;对KCl诱导收缩的最大抑制率分别为(79.36±11.66)%和(82.89±11.90)%.结论木脂素可能是南、北五味子发挥舒张气管平滑肌作用的主要活性成分. Objective To investigate the effects of lignans and polysaccharides of northern and southern Schis-andra chinensis on tension of isolated tracheal smooth muscle in rats.Method The lignans and polysaccharides of northern and southern Schisandra chinensis were prepared separately,and the lignans and polysaccharide content were determined by high performance liquid chromatography(HPLC)and phenol sulfuric acid method,respectively.In vitro tissue perfusion experiment was employed to analyze the effects of lignans and polysa-ccharides of northern and southern Schisandra chinensis on the resting state of tracheal rings,acetylcholine(ACh)and potassium chloride(KCl)precontraction of tracheal ring tension in rats.Results In the southern and northern Schisandra chinensis,the total contents of six lignans,Schisantherin A,Schizandrin A,Schizandrol A,Schizandrin B,Schizandrol B and Schizandrin C were(18.63±0.89)mg/g and(19.33±0.95)mg/g,respectively,and the contents of crude polysaccharides were 19.27 mg/g and 20.5 mg/g,respectively.No signi-ficant effects were observed on the tension of isolated rat tracheal rings at rest by lignans and polysaccharides(0.06~2.00 mg/mL)from both southern and northern Schisandra chinensis.No significant effect was also seen on ACh-precontracted tracheal ring tension by polysaccharide of southern and northern Schisandra chinensis,while lignan(0.25~1.75 mg/mL)dose dependently inhibited ACh-and KCl-induced tracheal ring contraction.The maximum inhibition rate of ACh-induced contraction was(68.21±10.08)%and(71.72±9.80)%,respecti-vely.The maximum inhibition rate of KCl-induced contraction was(79.36±11.66)%and(82.89±11.90)%,respectively.Conclusion Lignans may be the main active component in the diastole of tracheal smooth muscle exerted by both northern and southern Schisandra chinensis.
