帕金森病黑质基因的生物信息学分析

摘要 目的挖掘帕金森病黑质改变的关键信号分子,进一步阐明帕金森病发病的生物信息学基础。方法在GEO数据库中下载并筛选帕金森病黑质的差异表达基因,利用STRING在线数据库构建蛋白与蛋白互作网络分析,运用Cytoscape筛选出关键基因,通过DAVID数据库平台对差异基因进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析。结果下载得到基因芯片GSE20333的数据,获得63个差异基因,其中5个上调基因,58个下调基因。筛选出PIK3GG、P2RY10、LDHC、INADL等10个关键基因。GO分析显示差异基因主要富集在细胞外、细胞膜。KEGG分析主要设计催乳激素信号通路、造血细胞谱系和癌症中的胆碱代谢等信号通路。结论本研究得出的关键基因和信号通路可能与帕金森病黑质改变的分子过程有关,为深入研究帕金森病的治疗提供理论参考。 Objective To explore the key signal molecules related to altered substantia nigra in Parkin-son′s disease,and to further clarify the bioinformatics underlying onset of Parkinson′s disease.Methods From the GEO database,differentially expressed genes in substantia nigra of patients with Parkinson′s disease were downloaded and screened,and subjected to protein-protein interaction network construction and analysis using the STRING online database.The Cytoscape software was used to screen out key genes.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses of the differentially expressed genes were performed using the DAVID platform.Results The data of gene chip GSE20333 was downloaded and 63 differential genes were obtained,of which,5 were up-regulated genes and 58 were down-regulated.Ten key genes,including PIK3GG,P2RY10,LDHC,and INADL,were screened out.GO analysis showed that the differential genes were mainly enriched in the extracellular space and on cell membrane.KEGG analysis indicated that these genes mainly involved prolactin signaling pathway,hematopoietic cell lineage and choline metabolism in cancer.Conclusion The key genes and signal pathways identified in this study may be related to the molecu-lar processes related to altered substantia nigra in Parkinson′s disease.These findings provide the theoretical ref-erences for in-depth research on the treatment of Parkinson′s disease.
