黄精双向液体发酵的菌种优选及培养基优化研究 被引量:1

摘要 目的:以黄精为药性基质,以鸡腿菇、蛹虫草、秀珍菇为发酵菌种进行发酵,筛选出最适合于黄精液体发酵的菌种并优化其液体发酵培养基。方法:以抗氧化能力为指标筛选出黄精液体发酵的最优菌种,并以生物量为指标,通过单因素试验及正交试验筛选出最佳的液体发酵培养基。结果:鸡腿菇-黄精双向发酵后菌丝体得率为8.47 g/L(P<0.05),1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率为92.15%(P<0.01),羟自由基抑制能力为78.12 U/mL,总抗氧化能力为9.37 U/mL(P<0.01)。鸡腿菇发酵黄精最佳培养基为葡萄糖0%,麸皮含量5%,黄精质量浓度10 mg/mL,土豆20%,KH2PO40.1%,MgSO_(4)0.1%。按照该条件进行发酵、验证试验,菌丝体得率为(8.06±0.20)g/L。结论:为黄精双向液体发酵的研究奠定基础,并为黄精在保健品及化妆品中的应用提供一定的依据。 Objective:To select the most suitable strain for liquid fermentation and optimize the liquid fermentation medium with Huangjing(Polygonati Rhizoma)as medicinal substrate and mushroom,coprinus comatus,cordyceps militaris and pleurotus geesteranus as the fermentation strains.Methods:The optimal strains for the liquid fermentation of Huangjing was selected based on the antioxidant capacity as the index,and the best liquid fermentation medium was screened by single-factor test and orthogonal test with the biomass as the index.Results:the mycelial yield was 8.47 g/L(P<0.05),the 1,1-diphenyl-2-picylhydrazyl(DPPH)radical scavenging rate was 92.15%(P<0.01),the hydroxyl radical inhibitory capacity was 78.12 U/mL,and the total antioxidant capacity was 9.37 U/mL(P<0.01)after the bidirectional fermentation of coprinus comatus-Huangjing.The optimal medium for the fermentation of Huangjing by coprinus comatus was 0%glucose,5%bran content,10 mg/mL Huangjing mass concentration,20%potato,0.1%KH2PO4,and 0.1%MgSO_(4).According to the conditions of fermentation and verification test,mycelium yield was(8.06±0.20)g/L.Conclusion:This experiment lays the foundation for the study of bidirectional liquid fermentation of Huangjing and provides some basis for the application of Huangjing in health care products and cosmetics.
