抗原呈递相关基因在银屑病皮损中的表达研究

Expression of antigen presentation related genes in psoriasis lesions

目的探讨抗原呈递相关基因人类白细胞抗原(HLA)、内质网氨肽酶1(ERAP1)及CD1a在银屑病患者中的蛋白表达情况及其相关性。方法应用免疫组化方法对经临床及组织病理确诊为寻常型银屑病(PsV)38例、泛发性脓疱型银屑病(GPP)21例及健康对照组33例的皮损组织进行HLA-C、ERAP1、CD1a的蛋白表达检测,并进行统计学分析。结果PsV、GPP与健康对照组中HLA-C、ERAP1、CD1a均有蛋白表达。经检验,3种蛋白的阳性细胞比值在3组间差异具有统计学意义(P<0.05)。HLA-C的平均蛋白表达强度IOD值在PsV与对照组、PsV与GPP的组间比较中存在差异(P<0.05);ERAP1的平均蛋白表达强度IOD值在PsV与对照组、GPP与对照组的组间比较中存在差异(P<0.05);CD1a的平均蛋白表达强度IOD值在PsV与对照组的组间比较中存在差异(P<0.05)。利用Spearman相关在PsV组中进行分析,HLA-C与CD1a的表达具有相关性(rs=0.370,P=0.022);ERAP1与CD1a的表达具有相关性(rs=-0.383,P=0.018)。结论银屑病患者ERAP1、HLA-C、CD1a的蛋白在PsV皮损中呈高表达,并且具有相关性。ERAP1在GPP皮损中也呈高表达。3种抗原呈递基因可能在银屑病发病中具有重要作用。

Objective To investigate the protein expressions of antigen presenting related genes human leukocyte antigen(HLA),endoplasmic reticulum aminopeptidase 1(ERAP)and CD1a in patients with psoriasis and their correlation.Methods The application of immunohistochemical method for clinical and tissue pathological diagnosis of 38 cases of psoriasis vulgaris(PsV),21 cases of generalized pustular psoriasis(GPP)and 33 cases of health controls of skin lesions in groups performed protein expression testing of HLA-C、ERAP1 and CD1a,and conducted statistical analysis.Results HLA-C、ERAP1、CD1a were all expressed in PsV,GPP and healthy control group.The ratio of positive cells of the three proteins were significantly different among the three groups(P<0.05).The IOD value of average protein expression intensity of HLA-C was different between PsV and control group,PsV and GPP group(P<0.05);the IOD value of average protein expression intensity of ERAP1 was different between PsV and control group,GPP and control group(P<0.05);the IOD value of average protein expression intensity of CD1a was different between PsV and control group(P<0.05).Spearman regression was used to analyze the expression of HLA-C、ERAP1 and CD1a in PsV group.The expression of HLA-C and CD1a was correlated(r=0.370,P=0.022);the expression of ERAP1 and CD1a was correlated(r=-0.383,P=0.018).Conclusion In psoriasis patients,ERAP1、HLA-C、CD1a proteins were highly expressed in PsV lesions and were correlated.ERAP1 was also highly expressed in GPP lesions.Three antigen-presenting genes may play important roles in the pathogenesis of psoriasis.
