脂氧素A4保护大鼠脊髓缺血再灌注损伤的实验研究

摘要 探讨脂氧素A4通过抑制炎症对脊髓缺血再灌注损伤大鼠模型的保护作用。方法为:实验大鼠随机分为3组:脂氧素A4组(LXA4)、再灌注组(SCII组)和空白组(sham组),构建大鼠SCII模型,LXA4组和sham组大鼠在关腹后30 min应用10μL脂氧素A4(300 pmol)行脊髓鞘内注射。对3组大鼠在再灌注后6 h、12 h、24 h、48 h时间点时行BBB评分,并应用HE染色观察3组大鼠再灌注后24 h的脊髓组织性形态,分别在6 h、12 h、24 h、48 h测定同一节段脊髓组织MPO和TNF-α、IL-1β的活性。结果:与sham组相比,LXA4组和SCII组大鼠的BBB评分明显降低,而MPO、IL-1β及TNF-α活性明显升高,差异有显著意义(P>0.05)。LXA4组和SCII组两组间对比:(1)LXA4组大鼠在各个时间点BBB评分均明显高于SCII组,而MPO活性均明显降低(P<0.05);(2)LXA4组大鼠在12 h和24 h时TNF-α、IL-1β含量显著低于SCII组大鼠;(3)SCII组脊髓组织中较LXA4组出现大量炎症细胞浸润以及神经元变性坏死。结论:LXA4通过抑制炎症反应,从而起到治疗SCII的目的。 Objective:To investigate the protective effect of lipoxin A4 on rat spinal cord ischemia-reperfusion injury.Methods:Spinal cord ischemia-reperfusion injury models were built and randomly divided into sham group,ischemia-reperfusion group(SCII group)and experiment group(LXA4 group).LXA4 group rats were administered to get intrathecal injection of 10μl LXA4(300 pmol).Three groups of rats were scored by Basso-Beattie-Bresnahan(BBB)score at 6h,12h,24h,48h after reperfusion and were done HE staining at 24h after reperfusion.And then we measured the activity of myeloperoxidase(MPO)and IL-1β、TNF-αat 6h,12h,24h,48h after reperfusion.Results:Compared with sham group,SCII group and LXA4 group were significantly improved BBB scores,tumor necrosis factorα,interleukin-1βexpression and myeloperoxidase oxide activity content decreased(P>0.05).Compared with SCII group,(1)The BBB scores of LXA4 group were increased at 12h,24h,48h after reperfusion and the MPO activity of LXA4 group was decreased at each time(P<0.05);(2)The IL-1βand TNF-αof LXA4 group at 12h,48h after reperfusion were decreased significantly(P<0.05);(3)In the SCII group,there were a large number of degenerative neurons and infiltrated inflammatory cells.by comparison,the number of degenerative neurons in LXA4 group decreased and the nuclear form was restored to normal basically.Conclusion:LXA4 can achieve neural protection by inhibiting neuronal inflammation,thereby reducing spinal cord ischemia/reperfusion injury.
