细粒棘球绦虫表面抗原MKK1的生物信息学分析

摘要 目的运用生物信息预测网站及相关工具分析细粒棘球绦虫表面抗原MKK1的理化性质、抗原性等。方法EgMKK1氨基酸序列经NCBI数据库中下载,采用ProtParam分析蛋白的理化性质,SignalP-5分析信号肽,Euk-mPLoc 2.0进行亚细胞定位,ProtScale、SOSUI及DNASTAR分析亲疏水性,SOMPA分析二级结构,NetPhos分析磷酸化位点,MotifScan分析修饰位点,Swissmodel分析三级结构,TMHMM分析跨膜区域,ABCpred和IEDB预测B细胞抗原表位,SYFPEITHI预测T细胞抗原表位。结果EgMKK1由338氨基酸序列组成,分子式为C_(1688)H_(2687)N_(471)O_(497)S_(17),亲水指数为-0.167456,为亲水蛋白;无信号肽,含2个跨膜区,且定位于细胞质及细胞核中。二级结构中α螺旋占43.20%,β折叠占10.95%,β转角占4.14%,无规则卷曲占41.72%。EgMKK1能与HLA-A*02-01结合,且能被HLA-DRB*0401(DR4Dw4)分子呈递。结论生物信息学预测EgMKK1为亲水性蛋白,含有丰富的T、B细胞抗原表位,可为该蛋白的生物学功能研究及细粒棘球蚴病的防治研究提供参考。 Objective To explore the physicochemical properties and antigenicity of Echinococcus granulosus surface antigen MKK1 by using bioinformatics prediction website and related tools.Methods EgMKK1 amino acid sequences were downloaded from NCBI database and analyzed by ProtParam(property),SignalP-5(signal peptide),Euk-mPLoc 2.0(subcellular localization),ProtScale,SOSUI,DNASTAR(hydrophobicity),SOMPA(secondary structure),NetPhos(phosphorylation site),MotifScan(modification site),Swissmodel(tertiary structure)and TMHMM(transmembrane region).ABCpred and IEDB predicted B cell epitopes and SYFPEITHI predicted T cell epitopes.Results EgMKK1 is composed of 338 amino acid sequence,the molecular formula is C_(1688)H_(2687)N_(471)O_(497)S_(17),the hydrophilic index is-0.167456,it was a hydrophilic protein,no signal peptide,two transmembrane regions,and located in the cytoplasm and nucleus.In the secondary structure,αhelix 43.20%,βfold 10.95%,βcorner 4.14%,irregular crimp 41.72%.EgMKK1 has the ability to combine with HLA-A*02-01 and can be presented by HLA-DRB*0401(DR4Dw4)molecules.Conclusions Bioinformatics predicted that EgMKK1 was a hydrophilic protein.It contains rich T and B cell antigenic epitopes.It can provide reference for the study of the biological function of the protein and the prevention and treatment of E.granulosus.
