沙米总黄酮对1型糖尿病小鼠氧化应激的调节作用

摘要 目的研究沙米总黄酮(total flavonoids of Agriophyllum squarrosum,ATF)对1型糖尿病(type 1 diabetes mellitus,T1DM)小鼠糖脂代谢和氧化应激作用。方法采用单因素实验和响应面法优化ATF的最佳提取条件,研究ATF体外抗氧化活性及其对T1DM小鼠糖脂代谢和氧化应激作用。结果当乙醇浓度为83%、液固比34:1(mL/g)、提取时间65 min、提取温度50℃,ATF提取得率为(21.98±0.11)mg/g;ATF在体外表现出良好的抗氧化能力,ATF可显著降低T1DM小鼠总胆固醇(total cholesterol,TC)和甘油三酯(triglyceride,TG)水平,肝和肾脏系数显著降低,总超氧化物歧化酶(total superoxide dismutase,T-SOD)、谷胱甘肽(glutathione,GSH)和过氧化氢酶(catalase,CAT)活性显著升高,丙二醛(malonyldialdehyde,MDA)含量显著降低。结论ATF可有效改善小鼠糖脂代谢、器官系数和抗氧化酶,从而减少对肝组织的损伤。本研究结果可为沙米在开发辅助降糖功能食品和改善糖尿病方面的应用提供理论参考。 Objective To investigate the effects of total flavonoids of Agriophyllum squarrosum(ATF)on glucose and lipid metabolism and oxidative stress in type 1 diabetes mellitus(T1DM)mice.Methods The single factor experiment and response surface methodology were used to optimize the optimal extraction conditions of ATF,and the antioxidant activity of ATF in vitro and its effects on glucose and lipid metabolism and oxidative stress in T1DM mice were studied.Results When ethanol concentration was 83%,liquid-solid ratio was 34:1(mL/g),extraction time was 65 min,and extraction temperature was 50℃,the extraction yield of ATF was(21.98±0.11)mg/g;ATF showed good antioxidant capacity in vitro.ATF could significantly reduce the levels of total cholesterol(TC)and triglyceride(TG)in T1DM mice,significantly reduce the liver and kidney coefficients,and significantly reduce the total superoxide dismutase(T-SOD),glutathione(GSH)and catalase(CAT)activities,and significantly reduce the content of malondialdehyde(MDA).Conclusion ATF can effectively improve glucose and lipid metabolism,organ coefficient and antioxidant enzymes in mice,thus reducing the damage to liver tissue.The results of this study can provide a theoretical reference for the application of Agriophyllum squarrosum in the development of functional food for assisting in reducing blood sugar and improving diabetes.
