摘要
目的研究白芍总苷对系统性红斑狼疮小鼠肾脏损伤和肠道菌群的影响。方法将20只SPF级MRL/lpr雌性小鼠随机分为模型组、地塞米松组、白芍总苷低剂量组、白芍总苷高剂量组,每组5只。另取SPF级C57BL/6J雌性小鼠5只作为空白组。空白组及模型组小鼠给予生理盐水0.1 mL/10 g灌胃,地塞米松组给予地塞米松0.15 g/kg灌胃,白芍总苷低、高剂量组分别给予0.039 g/kg和0.078 g/kg白芍总苷混悬液灌胃,均1次/d,连续灌胃4周。取各组小鼠肾脏,HE染色观察肾脏病变情况,Masson染色观察肾间质纤维化情况;取各组小鼠结肠内容物进行16S rRNA测序,分析肠道菌群多样性及物种丰富度。结果HE染色和Masson染色显示,模型组肾小球系膜细胞和内皮细胞增生,肾间质有炎性浸润,肾小球明显萎缩,肾间质纤维化;与模型组比较,地塞米松组及白芍总苷低、高剂量组上述改变均明显减轻。肠道菌群方面:门水平上,白芍总苷低、高剂量组拟杆菌门(Bacteroidetes)相对丰度上调,厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)的相对丰度下调,其中白芍总苷低剂量组调节作用更明显;属水平上,白芍总苷高剂量组中乳杆菌属(lactobacillus)相对丰度表达较模型组上调,且产丁酸菌(butyricicoccus)表达富集。此外,白芍总苷可显著上调S-腺苷甲硫氨酸合成酶代谢途径表达。结论白芍总苷可改善系统性红斑狼疮小鼠肾损伤,其作用机制可能与上调肠道益生菌表达有关。
Objective It is to study the effects of total peony glycosides on kidney injury and intestinal flora in mice with systemic lupus erythematosus.Methods Twenty SPF-grade MRL/lpr female mice were randomly divided into model group,dexamethasone group,total peony glucoside low-dose group and total peony glucoside high-dose group,with 5 mice in each group.Five SPF-grade C57BL/6J female mice were also taken as the blank group.The mice in the blank group and model group were given normal saline 0.1 mL/10g by gavage,the ones in the dexamethasone group was given dexamethasone 0.15 g/kg by gavage,and the ones in the total peony glucoside low and high dose groups were given 0.039 g/kg and 0.078 g/kg total peony glucoside suspension by gavage respectively,all once/d,continuously treated for 4 weeks.The kidneys of the mice in each group were taken to observe the renal lesions by HE staining and observe the interstitial fibrosis by Masson staining;the colon contents of the mice in each group were sequenced with 16S rRNA to analyze the diversity of intestinal flora and species richness.Results HE staining and Masson staining showed that the glomerular thylakoid cells and endothelial cells in the model group were proliferated,there was inflammatory infiltration in the renal interstitium,glomerular atrophy and interstitial fibrosis;compared with the model group,the above changes were significantly reduced in the dexamethasone group and the low and high dose groups of total peony glucoside.The relative abundance of Bacteroidetes was up-regulated in the low and high dose groups,and the relative abundance of Firmicutes and Proteobacteria was down-regulated in the low and high dose groups,and the regulating effect was more obvious in the low dose group.The relative abundance of Lactobacillus genus(lactobacillus)expression in the group and the expression of butyric acid producing bacteria(butyricoccus)was enriched.In addition,total peony glycosides significantly up-regulated the expression of S-adenosylmethionine synthase metabolic pathway.Conclusion Total peony glycosides can improve renal injury in systemic lupus erythematosus mice,and its mechanism of action may be related to the upregulation of intestinal probiotic expression.
