基于公共数据库分析LDHA、LDHB基因在肺腺癌中的表达及其预后

摘要 目的探究乳酸脱氢酶A/B(LDHA/LDHB)在肺腺癌(LUAD)中的表达及其预后。方法使用R语言和韦恩图从基因表达谱交互式分析(GEPIA)数据库、UALCAN数据库和人类蛋白图谱(HPA)数据库中筛选出LUAD的潜在遗传生物标记物并验证,使用受试者工作特征(ROC)曲线、Kaplan-Meier生存曲线评估LDHA/LDHB在LUAD中的诊断及预后价值。通过富集分析LDHA/LDHB影响LUAD预后的潜在分子机制及与免疫细胞浸润之间的关系。结果LDHA和LDHB在LUAD组织中高表达,在正常组织中低表达;LDHA/LDHB表达与LUAD发生、进展及转移密切相关;ROC曲线分析表明,LDHA和LDHB在LUAD患者中具有较高的诊断价值;生存曲线显示,LDHA/LDHB表达较高的患者常存在预后不良;富集分析表明,LDHA和LDHB与LUAD的多条恶性肿瘤相关通路有关;LDHA和LDHB表达与免疫细胞浸润具有相关性(P<0.05),与Th2细胞呈显著正相关(P<0.05)。结论生物信息学综合分析表明,LDHA和LDHB在LUAD中表达增加,可能成为LUAD诊断和预后的潜在生物标志物和治疗靶点。 Objective To investigate the expression and prognostic value of lactate dehydrogenase A/B(LDHA/LDHB)in lung adenocarcinoma(LUAD).Methods Potential genetic biomarkers of LUAD were screened and validated from the Gene Expression Profiling Interactive Analysis(GEPIA)database,UALCAN database and Human Protein Atlas(HPA)database by using R software and the Wayne diagram.The diagnostic and prognostic value of LDHA/LDHB in LUAD was evaluated using receiver operating characteristic(ROC)curve and Kaplan Meier survival curve.The potential molecular mechanisms of LDHA/LDHB affecting the prognosis of LUAD,as well as the relationship between LDHA/LDHB and immune cell infiltration were analyzed by enrichment analysis.Results LDHA and LDHB were upregulated in LUAD when compared to normal tissues,indicating that the LDHA/LDHB expression was closely related to the occurrence,progression and metastasis of LUAD.ROC curve analysis showed that LDHA and LDHB had a high diagnostic value in patients with LUAD.The survival curve showed that patients with high LDHA/LDHB expression had a poor prognosis.Enrichment analysis revealed that LDHA and LDHB expression were related to several malignant tumor related pathways,as well as immune cell infiltration(P<0.05),especially that of Th2 cells(P<0.05).Conclusion Our comprehensive bioinformatics analysis revealed that the expression of LDHA and LDHB was increased in LUAD,suggesting that they could be useful biomarkers and therapeutic targets for the diagnosis and prognosis of LUAD.
