基于肿瘤微环境设计的高分子前药的研究进展

摘要 将药物改造成前药是修饰药物的一种常用手段,现已被广泛应用于抗肿瘤药物的设计中,其不仅可以改变母药溶解性,提高生物利用度,还能实现母药的靶向性。高分子前药作为前药设计的一个重要分支,既能发挥前药的优势,又能避免小分子药物在体内易被清除、选择性差等问题。笔者根据高分子前药的分类和设计原理,对近年来应用于抗肿瘤领域的靶向抗肿瘤高分子前药的研究情况和应用进行了综述。 Prodrug principle is a common means of modifying drugs,which has been widely used in the design of anti-tumor drugs.It can not only change the solubility and improve the bioavailability of parent drugs,but also achieve the targeted delivery of parent drugs.As an important branch of prodrug design,polymeric prodrugs can not only own the advantages of prodrugs,but also avoid some problems such as easy removal of small molecule drugs in vivo and poor selectivity.Based on the classification and design principles of polymer prodrugs,this paper reviews the research and application of targeted antitumor polymer prodrugs in the field of antitumors in recent years.
