FEM1B促进TRAIL诱导的HIV潜伏库细胞ACH-2的细胞凋亡

摘要 目的探究FEM1B对HIV潜伏库细胞ACH-2的凋亡影响及作用。方法利用慢病毒转染构建FEM1B过表达的ACH-2细胞株,采用FITC-Annexin-V/7-AAD染色检测FEM1B过表达的ACH-2细胞株的细胞自发凋亡以及TRAIL作用24 h后的细胞凋亡情况,以及用qPCR检测TRAIL作用前后细胞上清病毒载量。结果FEM1B过表达的细胞株的细胞自发凋亡没有显著变化,但在TRAIL作用下,过表达FEM1B的细胞株的凋亡率显著增加,同时FEM1B过表达后TRAIL作用下ACH-2的上清病毒载量明显上升。结论FEM1B能够促进TRAIL作用下HIV潜伏细胞的细胞凋亡和病毒载量。 Objective To study the role and effect of FEM1B in apoptosis of HIV reservoir ACH⁃2 cell lines.Methods A lentivirus used to overexpress FEM1B in ACH⁃2 cell lines.FITC⁃annexin⁃V/7⁃AAD staining was used to assess spontaneous cell apoptosis and cell apoptosis under TRAIL stimulation.Viral loads in culture supernatants were examined by qPCR.Results Spontaneous cell apoptosis remained unchanged in FEM1B⁃overexpressing cell lines.Cell apoptosis and viral loads in culture supernatants under TRAIL stimulation were significantly increased.Conclusions FEM1B increases TRAIL⁃mediated cell apoptosis and viral loads in culture supernatants of HIV reservoir cell lines.
