高压超短电场脉冲高选择性兔下腔静脉血管平滑肌细胞消融模型建立的探索

Establishment of a Rabbit Model for Selective Ablation of Inferior Vena Cava Vascular Smooth Muscle Cells Using High-Voltage Ultrashort Electric Field Pulses

摘要 目的高压超短电场脉冲通过瞬时、高能的局部电场变化,具有不破坏细胞完整结构、诱导细胞自主凋亡的生物学效应,这在抑制血管重构、管壁增生、血管狭窄方面具有潜在的应用价值。本研究自主研发出一款新型高压超短电场脉冲系统,探究该系统对兔下腔静脉作用后的生物学效应。方法利用1000 V/cm、1 Hz高压超短电场脉冲电刺激实验兔下腔静脉60次,常规饲养后,在术后第3天处死后,解剖分离实验兔下腔静脉血管标本,送HE病理染色常规检查。结果成功制备自主研发新型高压超短电场脉冲系统,高压超短电场脉冲作用点区域平滑肌凋亡,血管壁变薄,局部血管内皮细胞无明显损伤;无电刺激对照区域血管内皮细胞、平滑肌细胞及血管壁结构完整。结论高压超短电场脉冲,具有高选择性消融平滑肌细胞,保持周围血管内皮及平滑肌细胞、组织结构完整性的作用。 Objective High-voltage ultra-short electric field pulse has a biological effect of not destroying the intact structure of cells and inducing autonomous apoptosis of cells through instantaneous and high-energy local electric field changes,which has potential application value in inhibiting vascular remodeling,vessel wall proliferation and vascular stenosis.In this study,a new type of high-voltage ultra-short electric field pulse system was independently developed to explore the biological effects of the system on rabbit inferior vena cava.Methods The inferior vena cava(IVC)of experimental rabbits was stimulated by 1000 V/cm,1 Hz high voltage ultra-short electric field pulse for 60 times.After routine feeding,the IVC specimens of experimental rabbits were dissected and separated after death on the third day after surgery,and the specimens were sent to HE pathological staining for routine examination.Results The self-developed new high-voltage ultra-short electric field pulse system was successfully prepared.The smooth muscle was apoptosis,the vascular wall became thinner,and the local vascular endothelial cells had no obvious injury.In the control area without electrical stimulation,the vascular endothelial cells,smooth muscle cells and vascular wall structure were intact.Conclusion High pressure ultra-short electric field pulse can selectively ablation smooth muscle cells and maintain the integrity of peripheral vascular endothelium,smooth muscle cells and tissue structure.
