哮喘患儿IL-22、RAD50基因多态性与易感性的关系

摘要 目的探讨哮喘患儿白介素-22(IL-22)、RAD50基因多态性与疾病易感性的关系。方法选取2019年1月至2021年1月河南省平顶山市第一人民医院收治的哮喘患儿76例作为观察组,另选取同期体检健康的儿童62例作为对照组,收集两组背景资料,以聚合酶链式反应结合直接测序法检测IL-22(rs17224704位点、rs2227473位点)、RAD50(rs6871536位点、rs17166050位点)基因多态性,并对影响儿童哮喘的发生因素进行回归分析。结果两组出生方式、1岁内抗生素使用次数、饲养宠物、家族过敏史、家族哮喘史、被动吸烟等项目比较差异有统计学意义(P<0.05);两组IL-22基因rs17224704位点、rs2227473位点等位基因频率比较差异有统计学意义(P<0.05),RAD50基因rs6871536位点、rs17166050位点基因型、等位基因频率比较差异有统计学意义(P<0.05);rs17224704位点、rs2227473位点存在交互作用(P<0.05);Logistic回归分析显示,有家族过敏史、家族哮喘史儿童发生哮喘的风险增加(P<0.05),IL-22基因rs17224704位点携带等位基因A的儿童发生哮喘风险较携带T的儿童增加(P<0.05),RAD50基因rs6871536位点携带等位基因C的儿童发生哮喘风险较携带T的儿童增加(P<0.05)。结论IL-22基因rs17224704位点基因多态性、RAD50基因rs6871536位点基因多态性与儿童哮喘的易感性密切相关。 Objective To analyze the relationship between interleukin-22(IL-22)and RAD50 gene polymorphisms and suscep⁃tibility in children with asthma.Methods 76 children with asthma admitted to our hospital from January 2019 to January 2021 were se⁃lected as the observation group,and 62 healthy children who underwent physical examination during the same period were were selected as the control group.The background information of the two groups was collected.IL-22(rs17224704,rs2227473)and RAD50(rs6871536,rs17166050)gene polymorphisms were detected using polymerase chain reaction and direct sequencing.Regression analy⁃sis was performed to identify factors influencing the occurrence of childhood asthma.Results There were statistically significant differ⁃ences between the two groups in terms of birth method,antibiotic use frequency before the age of one,pet ownership,family history of al⁃lergies,family history of asthma,and passive smoking(P<0.05).The two groups showed statistically significant differences in the al⁃lele frequencies of rs17224704 and rs2227473 on IL-22 gene,as well as in the genotype and allele frequencies of rs6871536 and rs17166050 on RAD50 gene(P<0.05).There was an interaction effect between rs17224704 and rs2227473(P<0.05).Logistic re⁃gression analysis found that children with a family history of allergies or asthma had an increased risk of asthma(P<0.05).Children carrying the A allele at rs17224704 on IL-22 gene had an increased risk of asthma compared to those carrying the T allele(P<0.05),while children carrying the C allele at rs6871536 on RAD50 gene had an increased risk of asthma compared to those carrying the T al⁃lele(P<0.05).Conclusion The polymorphisms of rs17224704 on IL-22 gene and rs6871536 on RAD50 gene were closely associated with susceptibility to childhood asthma.
