卵巢癌类器官及重建肿瘤微环境的研究进展

摘要 卵巢癌是一种早期诊断率低、复发及耐药率高、具有较高致死率的妇科疾病。由于传统的卵巢癌细胞系和动物模型不能准确保留原肿瘤组织的组织学及基因特征,且建模效率低、时间长以及受到医学伦理约束等方面的局限性,这些传统模型尚不能满足卵巢癌的早期诊断和治疗领域的强烈需求。近年来,卵巢癌类器官模型能够高度还原原始肿瘤组成、形态发育、分子特征及基因信息,现已逐渐成为研究卵巢癌的新型研究模型工具,但是模拟卵巢癌的整个肿瘤微环境特征还是具有挑战性的。故本文对卵巢癌类器官的最新研究进展进行综述,并探讨重建卵巢癌微环境相关的动态共培养3D模型等优化模型,为探索卵巢癌的研究平台及治疗提供更多的助力。 Ovarian cancer(OC)is considered to be a gynecological disease with low early diagnosis rate,high recurrence and drug resistance rate,and high mortality rate.Because the traditional OC cell lines and animal models can’t accurately retain the histological and genetic characteristics of the original tumor tissues,and the limitations of low modeling efficiency,long time and medical ethics constraints,these traditional models can’t meet the strong demand in the field of early diagnosis and treatment of OC.In recent years,organoids models of OC can highly restore the original tumor composition,morphological development,molecular characteristics and genetic information,and have gradually become a new model tool for the study of OC.However,it is still challenging to simulate the characteristics of the entire tumor microenvironment of OC.Therefore,this study reviewed the latest research progress of OC organoids,and discussed the reconstruction of the dynamic co-culture 3D model related to the OC microenvironment and other optimization models,so as to provide more help for the exploration of the research platform and treatment of OC.
