干燥方式对小承气汤浸膏粉物理指纹图谱及化学成分的影响

摘要 目的考察4种干燥方式对小承气汤浸膏粉物理指纹图谱及化学成分含量的影响,优选合适的干燥方式。方法分别采用喷雾干燥、鼓风干燥、真空干燥、冷冻干燥制备小承气汤浸膏粉,以5个一级指标和9个二级指标构建物理指纹图谱,采用相似度评价、聚类分析(CA)比较其物理指纹图谱的差异,通过计算可压性参数比较4种浸膏粉的压缩成型性。采用HPLC法测定柚皮苷、新橙皮苷、芦荟大黄素、大黄酸、大黄素、和厚朴酚、厚朴酚、大黄酚、大黄素甲醚这9个成分的含量。结果4种干燥方式小承气汤浸膏粉物理指纹图谱相似度范围为0.865~0.950;聚类分析可将4种小承气汤浸膏粉分为2类;喷雾干燥粉具有良好的压缩成型性;4种干燥方式的小承气汤浸膏粉化学成分含量差异无统计学意义(P>0.05)。结论小承气汤采用喷雾干燥所得浸膏粉与采用冷冻干燥所得浸膏粉物理及化学性质基本一致,为该经典名方制剂工艺开发的干燥方式选择提供研究依据。 Objective To investigate the impacts of 4 drying methods on physical fingerprint and chemical composition content of Xiaochengqi Decoction extract powder,so as to select the appropriate drying method.Methods Xiaochengqi decoction extract powder was prepared by spray drying,blast drying,vacuum drying and freeze drying,respectively.Physical fingerprints were constructed with 5 primary indices and 9 secondary indices.The differences of physical fingerprints were compared by similarity evaluation and cluster analysis(CA).Compression formability of the 4 extract powders was compared by calculating compressibility parameters.The contents of 9 components,namely naringin,neohesperidin,aloe-emodin,rhein,emodin,honokiol,magnolol,chrysophanol and physcion,were determined by high performance liquid chromatography(HPLC).Results The similarity of physical fingerprints of Xiaochengqi Decoction extract powder with the 4 drying methods ranged from 0.865 to 0.950.The 4 categories of Xiaochengqi Decoction extract powder could be divided into 2 types by CA;Spray drying powder had good compression formability;There was no statistically significant difference in chemical composition content of Xiaochengqi Decoction extract powder by 4 drying methods(P>0.05).Conclusion The physical and chemical properties of the extract powder obtained by spray drying of Xiaochengqi Decoction are basically the same as those obtained by freeze drying.Therefore,it provides a research basis for the selection of drying methods for the development of this classic famous formula preparation process.
