高效液相色谱法测定黔产丽江麻黄中盐酸麻黄碱和盐酸伪麻黄碱的含量

Determination of Ephedrine Hydrochloride and Pseudoephedrine Hydrochloride in Lijiang Ephedra from Guizhou by HPLC

摘要 目的 建立高效液相色谱法(HPLC)测定黔产丽江麻黄中盐酸麻黄碱和盐酸伪麻黄碱的含量,为黔产丽江麻黄的质量评价提供方法。方法 采用C_(18)色谱柱,以乙腈-0.1%磷酸溶液(含0.1%三乙胺)为流动相,梯度洗脱,流速:1.0 ml/min,检测波长为206 nm。结果 盐酸麻黄碱线性方程:Y=2.364 3×10^(6)X-4.029 3×10^(4),R=1;盐酸伪麻黄碱线性方程:Y=2.417 78×10^(6)X-4.978 7×10^(4),R=1;线性范围为0.120 2~2.404 0μg和0.117 2~2.334 1μg;平均加样回收率(n=6)分别为100.28%(RSD=0.67%)和100.78%(RSD=0.69%)。结论 所建立的方法简便易行,重复性好,可用于黔产丽江麻黄的含量测定和质量评价。 Objective To establish a high performance liquid chromatography(HPLC) method for the determination of ephedrine hydrochloride and pseudoephedrine hydrochloride in Lijiang ephedra produced in Guizhou, and provide a method for the quality evaluation of Lijiang ephedra produced in Guizhou. Methods C_(18) column with acetonitrile-0.1% phosphoric acid solution(containing 0.1% triethylamine) as mobile phase was used for gradient elution;the flow rate was 1.0 ml/min;the detection wavelength was 206 nm. Results The linear range of ephedrine hydrochloride was Y=2.364 3×10^(6)X-4.029 3×10^(4), R=1;and range of linearity was 0.120 2~2.404 0 μg;the regression equation of pseudoephedrine hydrochloride was Y=2.417 8×10^(6)X-4.978 7×10^(4), R=1;and range of linearity was 0.117 2~2.334 1 μg. The average recoveries were 100.28%(RSD=0.67%) and 100.78%(RSD=0.69%). Conclusion The method is accurate, simple and reproducible, so it can be used toContent determination and evaluate the quality of Lijiang ephedra in Guizhou.
