摘要
目的探讨复元醒脑汤对大鼠局灶性脑缺血损伤后血管新生的影响及可能机制。方法60只健康雄性(SPF级)SD大鼠制备大脑中动脉闭塞(MCAO)模型,随机分为模型组、复元醒脑汤组、尼莫地平组,另设10只假手术组。术后2小时、第3天、第7天进行Longa评分;HE染色观察脑梗死病理形态;免疫荧光检测大脑缺血半暗带区皮质Ki67表达;免疫组化检测血管内皮生长因子A(VEGFA)、血管内皮生长因子受体1(VEGFR1)蛋白的表达;ELISA检测脑源性神经营养因子(BDNF)蛋白的表达。结果与假手术组比较,模型组神经功能评分显著升高(P<0.01),大脑缺血半暗带区皮质Ki67、VEGFA、VEGFR1蛋白表达增多(P<0.05或P<0.01),BDNF表达下降(P<0.05);与模型组相比,复元醒脑汤组Longza评分下降(P<0.05),大脑缺血半暗带区皮质Ki67、VEGFA、VEGFR1、BDNF蛋白表达均增多(P<0.05或P<0.01)。结论复元醒脑汤可改善神经功能缺损症状,上调Ki67、VEGFA、VEGFR1和BDNF蛋白表达,促进脑缺血后血管新生,发挥脑保护作用。
Objective To investigate the effects and possible mechanisms of Fuyuan Xingnao Decoction on angiogenesis after focal cerebral ischemia in rats.Methods 60 healthy male(SPF grade)SD rats with middle cerebral artery occlusion(MCAO)model were randomly divided into model group,Fuyuan Xingnao group and Nimodipine group,and another 10 sham-operated groups.Longa score was performed at 2 hours,day 3 and day 7 postoperatively;The pathological changes of cerebral infarction were ob-served by HE staining.The expression of Ki67 protein in the cerebral ischemic penumbra was detected by immunofluorescence;The expression of vascular endothelial growth factor A(VEGFA)and vascular endothelial growth factor receptor 1(VEGFR1)protein was detected by immunohistochemistry;The expression of brain-derived neurotrophic factor(BDNF)was detected by ELISA.Results Compared with the sham-operated group,Neurological function scores was significantly higher in the model group(P<0.01),and the expression of Ki67,VEGFA and VEGFR1 protein in the cortex of ischemic penumbra was increased(P<0.05 or P<0.01),while the expression of BDNF was decreased(P<0.05).compared with the model group,the Lon-ga score decreased in the Fuyuan Xingnao group(P<0.05),and the expression of Ki67,VEGFA,VEGFR1 and BDNF in the cortex of ischemic penumbra was increased(P<0.05 or P<0.01).Conclusion Fuyuan Xingnao Decoction can improve the symptoms of neurological deficits,up-regulate the expression of Ki67,VEGFA,VEGFR1 and BDNF proteins,promote angio-genesis after cerebral ischemia,and play a protective role in the brain.
