基于RNA-Seq数据分析油酸对牛前体脂肪细胞分化及PPAR信号通路的影响 被引量:1

Effects of Oleic Acid on Differentiation of Bovine Precursor Adipo⁃cytes and PPAR Signaling Pathway Based on RNA-Seq Data

摘要 利用100μmol/L油酸对细胞进行诱导分化,通过RNA-Seq技术对分化后48,96 h的细胞进行转录组测序,对差异表达基因进行GO功能注释和KEGG富集分析。结果表明:与分化48 h时相比,分化96 h时差异表达基因数增加3519个,差异表达基因的分子功能、参与的生物过程和细胞组成随分化时间不同均发生变化,生化代谢途径和信号转导途径也差异较大;由过氧化物酶体增殖物激活受体(Peroxisome proliferatorsactivated receptors,PPARs)信号通路的分析结果可以得出,分化48 h时,PPARγ的表达上调,信号通路成员PLIN2、CPT1β,SREBP1、FABP4上调,SCD表达下调;分化96 h时,PLIN2、SREBP1、CPT1β显著上调,PPARγ、SCD的表达显著下调,FABP4表达无显著变化。说明油酸对延边牛前体脂肪细胞分化过程中的转录组学及信号通路存在一定影响。 Cell differentiate was induced with 100μmol/L oleic acid,and transcriptome sequencing was performed on cells by RNA-Seq technology at 48 and 96 h after differentiation.GO functional annotation and KEGG enrichment analysis were performed for differentially expressed genes.The results showed that the number of differentially expressed genes increased by 3519 at 96 h compared with that at 48 h.The molecular functions,biological processes and cell composition of differentially expressed genes changed with differentiation time.Biochemical metabolic pathways and signal transduction pathways were also different.According to the analysis results of PPAR signaling pathway,PPARγexpression was up-regulated at 48 h differentiation,while the expression of SCD was down-regulated in PLIN2,CPT1β,SREBP1 and FABP4.At 96 h of differentiation,the expression of PLIN2,SREBP1 and CPT1βwere significantly up-regulated,PPARγand SCD were down-regulated,and FABP4 expression was not significantly changed.Therefore,the effects of oleic acid on transcriptome and signal pathway of Yanbian cattle preadipocytes were different at different differentiation time.
