摘要
为了解中华绒螯蟹不同群体的遗传结构,利用微卫星分子标记,分析中华绒螯蟹单年系F5代选育群体、“长江2号”和长江野生群体的遗传多样性。结果表明,6个微卫星位点的等位基因数为7~11,有效等位基因数为4.5394~9.5294,观测杂合度为0.6389~0.8611,期望杂合度为0.7907~0.9077,多态信息含量为0.7797~0.8951,6个微卫星位点均具有高度多态性。3个河蟹群体的期望杂合度为0.8176~0.8478,多态信息含量为0.7841~0.8125,表明所有群体均具有高度遗传多样性。遗传分化指数值为0.05269~0.08482,表明所有群体间均有不同程度的遗传分化。AMOVA分析显示,群体间变异占总变异的5.34%,群体内个体间的变异占总变异的94.66%。基于Nei氏遗传距离构建的UPGMA系统进化树显示,单年系F5代选育群体先与“长江2号”群体聚为一类,再与长江野生群体聚为一类。
In order to explore the genetic structure of Chinese Mitten Crab(Eriocheir sinensis)in different populations,six microsatellites with polymorphism are selected from the reported microsatellites,and the genetic diversity of three Chinese mitten crab populations.“Yangtze River No.3”,“Yangtze River No.2”and Yangtze River wild population,were analyzed.The results showed that the number of alleles of the six microsatellite loci was 7-11,the number of effective alleles was 4.5394-9.5294,the observed heterozygosity was 0.6389-0.8611,and the expected heterozygosity ranged from 0.7907 to 0.9077,polymorphic information content ranged from 0.7797 to 0.8951,and the six microsatellite loci were highly polymorphic.The expected heterozygosity of the three populations ranges from 0.8176 to 0.8478,and the polymorphic information content range from 0.7841 to 0.8125,indicating that all populations have high genetic diversity,and the level of genetic diversity is“Yangtze River No.2>Yangtze River No.3>Yangtze River wild population”.The genetic differentiation index ranges from 0.05269 to 0.08482,indicating that all populations have different degrees of genetic differentiation.AMOVA analysis show that the variation among populations account for 5.34%of the total variation,the variation among individuals within the population account for 94.66%of the total variation.The UPGMA phylogenetic tree constructed based on Nei’s genetic distance show that the“Yangtze River No.3”population is first clustered with the“Yangtze River No.2”population,and then clustered with the wild population of the Yangtze River.
