小鼠肺纤维化CT影像自动分析方法

Automated analysis method of CT images for mice pulmonary fibrosis

摘要 目的基于CTAN软件建立小鼠肺纤维化(PF)CT影像自动分析方法并观察其自动分析效果。方法将15只雄性c57小鼠随机分为博莱霉素组(A组)、博莱霉素+尼达尼布组(B组)及对照组(C组),每组5只。于建模第1、5、8、11及15天对A、B组采用腹腔注射博莱霉素构建PF模型,对C组以相同方法注射等量生理盐水;于第25、28、31及34天对B组腹腔注射尼达尼布以抑制PF。于第41天对所有小鼠行胸部CT扫描,并将图像导入CTAN软件,通过编辑和优化程序建立自动分析方法,提取肺CT影像区域,构建3D模型并进行数据分析;之后处死小鼠,取肺组织行Masson染色,观察肺部胶原沉积,评价PF Ashcroft评分,并检测肺羟脯氨酸含量,分析PF CT影像自动分析结果与Ashcroft评分及肺羟脯氨酸含量的一致性。结果A组肺组织区域体积与肺区域体积比及肺区域CT值均高于C组(t=14.06、10.32,P均<0.01)和B组(t=11.22、8.29,P均<0.01);其肺羟脯氨酸含量和Ashcroft评分亦均高于C组(t=11.92、23.39,P均<0.01)和B组(t=7.44、12.07,P均<0.01)。以小鼠PF CT影像自动分析方法获得的肺组织区域体积与肺区域体积比与PF Ashcroft评分和肺羟脯氨酸含量(R2=0.9455、0.8450,P均<0.001),以及小鼠肺区域CT值与PF Ashcroft评分和肺羟脯氨酸含量的一致性均佳(R2=0.8950、0.8480,P均<0.001)。结论所构建的CT影像自动分析方法用于分析小鼠PF效果良好。 Objective To establish an automatic analysis method of CT images for mice pulmonary fibrosis(PF)based on CTAN software,and to explore its application value.Methods Fifteen male c57 mice were randomly divided into Bleomycin group(group A),Bleoycin+Nitedanib group(group B)or control group(group C)(each n=5).At the 1st,5th,8th,11th and 15th day of modeling,PF model was established through intraperitoneal injection of Bleomycin in group A and B,while equal dose of normal saline was injected with the same method in group C.At the 25th,28th,31st and 34th day of modeling,mice in group B were given intraperitoneal injection of Nidanib to inhibit PF.Chest CT scanning was performed on the 41st day of modeling for all 15 mice,then the images were imported into CTAN software,and an automatic analysis method was established by editing and optimizing procedures to achieve lung CT image region extraction,3D model construction and data analysis.Then the mice were sacrificed,and the lung tissue were taken for Masson staining,the deposition of lung collagen was observed to evaluate PF Ashcroft score.Meanwhile,the lung hydroxyproline content were detected.The consistencies of automatic analysis results of CT images for mice PF with Ashcroft score and the results of lung hydroxyproline content were analyzed.Results The ratio of lung tissue area volume to lung area volume and CT value of lung area in group A were higher than those in group C(t=14.06,10.32,both P<0.01)and B(t=11.22,8.29,both P<0.01).The lung hydroxyproline content and Ashcroft score in group A were both higher than those in group C(t=11.92,23.39,both P<0.01)and B(t=7.44,12.07,both P<0.01).Good agreement of the ratio of lung tissue area volume to lung area volume obtained with this automatic analysis method was found with PF Ashcroft score,lung hydroxyproline content(R 2=0.9455,0.8450,both P<0.001),also between CT value of lung area with PF Ashcroft score and lung hydroxyproline content in mice(R 2=0.8950,0.8480,both P<0.001).Conclusion The established automatic analysis method of mice CT images had good application value for exploring mice PF.
