血清CCL11、PRDX1水平与炎症性肠病患者肠道菌群、肠黏膜屏障功能的相关性 被引量:6

Correlation between serum CCL11 and PRDX1 levels with intestinal microflora and intestinal mucosal barrier function in patients with inflammatory bowel disease

摘要 目的探究炎症性肠病(inflammatory bowel disease,IBD)患者血清CC趋化因子配体11(CC chemokine ligand 11,CCL11)、过氧化物还原蛋白1(peroxiredoxin 1,PRDX1)的表达水平与肠道菌群以及肠黏膜屏障功能的相关性。方法选取2019年3月至2022年3月我院收治的168例IBD患者作为IBD组,同期正常健康体检者120名作为对照组;应用qRT-PCR法检测血清中CCL11、PRDX1表达水平;通过半自动微生物鉴定系统进行鉴定,并计算粪便中肠道菌群数量;采用ELISA、鲎试验、可见分光光度法分别检测D-乳酸、内毒素、二胺氧化酶(diamine oxidase,DAO)水平;采用Pearson相关性分析血清CCL11、PRDX1表达水平相关性以及二者与肠道菌群数量、肠黏膜屏障功能的相关性。结果IBD组的血清中CCL11、PRDX1水平、CDAI评分和Mayo评分显著高于对照组(P<0.05),CCL11与PRDX1呈正相关(P<0.05),CCL11、PRDX1与CDAI评分、Mayo评分之间呈正相关(P<0.05);IBD组肠球菌、大肠杆菌的数量显著高于对照组,双歧杆菌、类杆菌数量显著低于对照组(P<0.05);IBD患者血清CCL11、PRDX1的表达水平与肠球菌、大肠杆菌数量呈正相关,与双歧杆菌、类杆菌数量呈负相关(P<0.05);IBD组D-乳酸、内毒素、DAO的含量显著高于对照组(P<0.05);IBD患者血清CCL11和PRDX1的表达水平与D-乳酸、内毒素、DAO的含量呈正相关(P<0.05)。结论IBD患者血清CCL11、PRDX1表达水平显著升高,二者表达呈正相关,且与肠道菌群以及肠黏膜屏障功能具有一定的相关性。 Objective To explore the correlation between the expression levels of serum CC chemokine ligand 11(CCL11)and peroxiredoxin 1(PRDX1)with the intestinal microflora and intestinal mucosal barrier function in patients with inflammatory bowel disease(IBD).Methods From Mar.2019 to Mar.2022,168 patients with IBD who were admitted to our hospital were regarded as the IBD group,and 120 people with normal physical examinations were regarded as the control group;the expression levels of CCL11 and PRDX1 in serum were detected by qRT-PCR;semi-automatic microbial identification system was applied to identify,and the number of intestinal microflora in feces was calculated;the levels of D-lactate,endotoxin and diamine oxidase(DAO)were detected by ELISA,Limulus test and visible spectrophotometry respectively.Pearson correlation was used to analyze the correlation of serum CCL11 and PRDX1 expression levels and their correlation with the number of intestinal microflora and intestinal mucosal barrier function.Results The expression levels of serum CCL11,PRDX1,CDAI score and Mayo score in the IBD group were greatly higher than those in the control group(P<0.05),and the expression level of CCL11 was positively correlated with PRDX1(P<0.05).There was a positive correlation between the expression level of CCL11,PRDX1 and CDAI,Mayo score;the numbers of Enterococcus and Escherichia coli in the IBD group were greatly higher than those in the control group,and the numbers of Bifidobacterium and Bacteroides were greatly lower than those in the control group(P<0.05);the expression levels of serum CCL11 and PRDX1 in IBD patients were positively correlated with the numbers of Enterococcus and Escherichia coli,and negatively correlated with the numbers of Bifidobacterium and Bacteroides(P<0.05);the contents of D-lactate acid,endotoxin and DAO in the IBD group were greatly higher than those in the control group(P<0.05);the expression levels of serum CCL11 and PRDX1 in IBD patients were positively correlated with the contents of D-lactate,endotoxin and DAO(P<0.05).Conclusion The expression levels of serum CCL11 and PRDX1 in IBD patients are greatly increased,the expressions of the two are positively correlated,and have a certain correlation with the intestinal microflora and intestinal mucosal barrier function.
