摘要
目的 本文研究了试剂盒抗原单组份不稳定性的影响因素;方法 通过抗原缓冲液离子强度、pH、不同试剂瓶材质和添加阻断剂等方法,测定不同条件的抗原溶液37℃加速后发光值的降幅,判断抗原不稳定的主要原因。结果 在抗原溶液中加入高分子聚合物泊洛沙姆12600、14600,抗原37℃热加速10 d后,稳定性得到明显的提升,发光值降幅由60%降低至8%。结论 高分子聚合物对抗原聚集有抑制作用,维持抗原结构的稳定。
【Objective】 To study the influencing factors of the instability of the single component of the antigen in the kit.【Methods】 Through the methods of antigen buffer ionic strength,pH,different reagent bottle materials and adding blocking agents,the antigen solutions under different conditions were determined at 37℃,the decrease of the luminescence value after acceleration was used to determine the main reason for the instability of the antigen.【Results】 After adding the high molecular polymer poloxamer 12600 and 14600 to the antigen solution,after the antigen was heated at 37°C for 10 days,the stability was significantly improved,and the decrease of luminescence value decreased from 60% to 8%.【Conclusion】 The macromolecular polymer has an inhibitory effect on antigen aggregation and maintains the stability of the antigen structure.
