瑞马唑仑诱导PC-12细胞炎症因子表达和凋亡 被引量:1

摘要 目的 探讨瑞马唑仑对大鼠肾上腺嗜铬细胞瘤细胞PC-12炎症因子表达和凋亡的影响。方法 不同浓度的瑞马唑仑(25、50、125、250μmol/L)处理PC-12细胞,并设置对照组,细胞活力检测试剂盒(CCK-8)检测细胞增殖,酶联免疫吸附测定(ELISA)检测肿瘤坏死因子α (TNF-α)、白细胞介素-6 (IL-6)、白细胞介素-1β (IL-1β)的水平,流式细胞术检测细胞凋亡,Western blot检测凋亡蛋白Bcl-2和Bax的表达。结果 与对照组相比,瑞马唑仑处理组的细胞增殖能力、Bcl-2蛋白表达水平明显降低(P<0.05),而炎性因子TNF-α、IL-6、IL-1β水平、细胞凋亡率、Bax蛋白表达水平明显升高(P<0.05)。结论 瑞马唑仑可诱导PC-12细胞的炎症反应,并能通过调节凋亡相关蛋白的表达促进PC-12凋亡。 【Objective】 To investigate the effect of remazolam on the expression of inflammatory cytokines and apoptosis of rat adrenal pheochromocytoma cells PC-12.【Methods】 PC-12 cells were treated with different concentrations of remazolam(25,50,125,250 μmol/L),and the control group was set up.Cell proliferation was detected by CCK8,the levels of TNF-α,IL-6,IL-1βwere detected by ELISA,cell apoptosis was detected by flow cytometry,and the expressions of apoptosis proteins Bcl-2 and Bax were detected by Western blot.【Results】 Compared with control group,the cell proliferation ability and Bcl-2 protein expression were significantly decreased(P<0.05),while the levels of inflammatory factors TNF-α,IL-6,IL-1β,apoptosis rate and Bax protein expression were significantly increased in the remazolam treatment group(P<0.05).【Conclusion】 Remazolam can induce the inflammatory response of PC-12 cells and promote the apoptosis of PC-12 cells by regulating the expression of apoptosis-related proteins.
