基于补益脾肾研究独活寄生贴对佐剂性关节炎大鼠的治疗作用

Effect of Duhuo Jisheng paste on rats with adjuvant arthritis based on invigorating the spleen and kidney

目的研究独活寄生贴对佐剂性关节炎大鼠的治疗作用。方法采用大鼠右后足部注射完全弗氏佐剂,建立佐剂性关节炎模型。实验分为正常对照组、模型组、穴位贴组、阳性组,在“脾俞”“肾俞”给予独活寄生贴。观察各组大鼠足肿胀程度;记录大鼠体质量;测定足关节炎指数评分;苏木素-伊红染色检测大鼠踝关节病理形态变化;利用酶联免疫吸附测定法检测大鼠血清中IL-1β、IL-10的表达水平;摘取大鼠脾脏、胸腺,计算脾脏、胸腺指数。同时取小鼠,在“脾俞”“肾俞”给予独活寄生贴进行热板致痛实验,观察该穴位贴的镇痛作用。结果与正常对照组比较,模型组大鼠体质量自造模第9日起显著下降(P<0.01),足肿胀度及关节指数评分显著升高(P<0.01),踝关节形态功能受损,IL-1β水平显著升高(P<0.01),IL-10水平显著下降(P<0.01),胸腺、脾脏指数升高(P<0.05)。与模型组比较,阳性对照组和治疗组大鼠踝关节滑膜组织病理学改善明显,自造模第9日起体质量升高(P<0.05),足肿胀度、关节炎指数以及胸腺、脾脏指数降低(P<0.05),血清炎症因子IL-1β水平显著降低(P<0.01),抗炎因子IL-10水平显著升高(P<0.01)。结论独活寄生贴可以降低大鼠的炎症反应,改善机体功能,从而发挥治疗佐剂性关节炎的作用。

Objective To determine the therapeutic effect of Duhuo Jisheng paste on rats with adjuvant arthritis.Methods Complete Freund’s adjuvant(CFA)was injected into the right hind foot of rats to establish the adjuvant arthritis model.The experiment was divided into a normal control group,a model group,a acupoint paste group and a positive group.The spleen and the kidney acupoints were given Duhuo Jisheng paste.The degree of foot swelling was observed.The body mass of the rats was recorded.The foot arthritis index score was measured.Hematoxylin-eosin staining was used to detect the pathological changes of the ankle joint of the rats.The levels of IL-1βand IL-10 in the serum were detected by ELISA.The spleen and thymus of the rats were removed and their index was calculated.At the same time,mice were given Duhuo Jisheng paste in spleen and kidney acupoints for hot plate pain test to observe the analgesic effect of the acupoint paste.Results Compared with the normal control group,the body mass of the model group was significantly decreased from day 9(P<0.01),the foot swelling degree and joint index score were significantly increased(P<0.01),the ankle joint morphology and function were impaired,the serum inflammatory factor IL-1βlevel was significantly increased(P<0.01),and the anti-inflammatory factor IL-10 level was significantly decreased(P<0.01).The indexes of thymus and the spleen were increased from day 9(P<0.05).Compared with the model group,the pathological changes of the ankle synovium in the positive group and the treatment group were significantly improved,the body mass was increased(P<0.05),the toe volume,arthritis indexes,thymus and spleen index were decreased(P<0.05),and the level of serum inflammatory factor IL-1βwas significantly decreased(P<0.01).The level of anti-inflammatory factor IL-10 was significantly increased(P<0.01).Conclusion Duhuo Jisheng paste can reduce the inflammatory response,improve the body function in rats,and treat adjuvant arthritis.
