摘要
目的:探究海洋鱼低聚肽螯合铁(MCOP-Fe)的小肠吸收特性,并与传统补铁剂硫酸亚铁(FeSO_(4))进行比较。方法:建立体外大鼠外翻肠囊模型,选取雌性Sprague Dawley大鼠40只,随机分为8组【低、中、高剂量(3,5,10μg/mL)MCOP-Fe组;低、中、高(3,5,10μg/mL)FeSO_(4)组;10μg/mL MCOP-Fe+500μg/mL植酸组;10μg/mL FeSO_(4)+500μg/mL植酸组】,用肠道单位面积铁累计吸收率Q评价不同肠段吸收铁的效力。结果:1)中、低、高浓度MCOP-Fe组在十二指肠、空肠及回肠的Q值随浓度呈现梯度增加,而FeSO_(4)组没有类似的趋势;2)中、低浓度FeSO_(4)组的Q值几乎都有高于MCOP-Fe组的趋势,而在高浓度时相反;3)添加500μg/mL植酸后,高浓度MCOP-Fe组和FeSO_(4)组Q值均有下降,高浓度FeSO_(4)组下降更为显著(P<0.05)。结论:在3~10μg/mL剂量范围内,FeSO_(4)的小肠吸收率呈现饱和的平衡趋势,而MCOP-Fe呈现梯度上升的趋势;同时MCOP-Fe受植酸的抑制作用小,相对FeSO_(4)更加稳定,应用前景良好。
Objective:To explore the absorption characteristics of marine fish oligopeptide chelated iron(MCOP-Fe)in small intestine,and compare it with the traditional iron supplement-ferrous sulfate(FeSO_(4)).Methods:40 female Sprague Dawley rats were randomly divided into 8 groups(L-MCOP-Fe:3μg/mL MCOP-Fe;M-MCOP-Fe:5μg/mL MCOP-Fe;H-MCOP-Fe:10μg/mL MCOP-Fe;L-FeSO_(4):3μg/mL FeSO_(4);M-FeSO_(4):5μg/mL FeSO_(4);H-FeSO_(4):10μg/mL FeSO_(4);H-MCOP-Fe+PA:10μg/mL MCOP-Fe+500μg/mL phytic acid;H-FeSO_(4)+PA:10μg/mL FeSO_(4)+500μg/mL phytic acid).To establish the model of everted gut sac in vitro,the effect of iron absorption in different intestinal segments was evaluated by the cumulative iron absorption rate per unit area(Q).Results:1)The Q values of duodenum,jejunum and ileum in three MCOP-Fe groups increased with concentration gradient,but there was no similar trend in FeSO_(4)groups;2)The Q values of group L-FeSO_(4)and M-FeSO_(4)had a tendency to be higher than those of group L-MCOP-Fe and M-MCOP-Fe,but were opposite at high concentration(10μg/mL);3)After adding 500μg/mL phytic acid,the Q values of groups H-MCOP-Fe+PA and H-FeSO_(4)+PA decreased compared with groups H-MCOP-Fe and H-FeSO_(4),and the decrease degree of group H-FeSO_(4)+PA was more significant(P<0.05).Conclusion:In the range of 3-10μg/mL,the absorption rate of FeSO_(4)in small intestine showed an equilibrium trend of saturation,while the MCOP-Fe showed a gradient trend of increase;at the same time,MCOP-Fe was less inhibited by phytic acid,and was more stable than FeSO_(4),which has good application prospects.
