摘要
通过观察白藜芦醇对镧暴露仔鼠皮层神经细胞caspase-3表达的影响,探讨其对镧暴露仔鼠的神经保护作用。建立白藜芦醇干预镧暴露仔鼠模型,用HE染色法观察皮质神经细胞形态,用实时定量PCR法检测皮质组织caspase-3 mRNA表达水平,用Western-blot法检测皮质组织caspase-3蛋白表达水平。HE染色结果显示:阴性对照组细胞数量多,形态正常;阳性对照组细胞数量最少,细胞损伤明显;随着白藜芦醇剂量的增加,干预组仔鼠皮质神经细胞损伤情况逐渐得到改善。实时定量PCR结果显示:仔鼠皮质组织caspase-3 mRNA表达在各剂量白藜芦醇干预组之间出现差异,与阳性对照组相比,中、高剂量白藜芦醇干预组仔鼠皮质组织中caspase-3 mRNA表达量明显降低,差异有统计学意义(P﹤0.05)。Western-blot实验结果显示:高剂量白藜芦醇干预组caspase-3蛋白表达水平明显低于阳性对照组和低剂量白藜芦醇干预组,差异有统计学意义(P﹤0.05)。研究结果提示白藜芦醇可改善镧暴露导致的仔鼠大脑皮质发育损伤,这可能与皮质组织caspase-3的mRNA和蛋白表达水平降低有关。
By observing the effect of resveratrol on the expression of caspase-3 in cortical nerve cells of lanthanum-exposed rats,the neuroprotective effect of resveratrol on lanthanum-exposed rats was discussed.The lanthanum-exposed rat model with resveratrol intervention was established.The morphology of cortical nerve cells was observed by HE staining.The caspase-3 mRNA expression in cortical tissue was detected by real-time quantitative PCR.The caspase-3 protein expression in cortical tissue was detected by Western-blot.HE staining results showed that the negative control group had a large number of cells with normal morphology;the number of cells in the positive control group was the least,and the cell damage was obvious.With the increase of resveratrol dose,the damage of cortical nerve cells was gradually improved.The results of real-time quantitative PCR showed that the caspase-3 mRNA expression in the cortical tissue of offspring rats was different among the resveratrol intervention groups.Compared with the positive control group,the caspase-3 mRNA expression in the cortical issue of the offspring rats in the middle and high dose resveratrol intervention groups was significantly decreased,and the difference was statistically significant(P<0.05).Western-blot results showed that the caspase-3 protein expression level in the high-dose resveratrol intervention group was significantly lower than that in the positive control group and the low-dose resveratrol intervention group(P﹤0.05).The results suggest that resveratrol can improve the developmental damage of cerebral nerve cells in offspring rats caused by lanthanum exposure,which may be related to the decrease of caspase-3 mRNA and protein expression in cortical tissue.
