摘要 为分析lncRNA-MSTRG.13836.1的功能,确定杜泊羊毛囊周期性生长发育相关的关键靶向关系对,选择8只2周岁杜泊羊母羊作为研究对象,其中5只脱毛羊为试验组,另3只不脱毛羊为对照组。在3个时间点收集它们的皮肤样本并进行RNA测序(RNA-seq),通过lncRNA差异表达分析筛选关键lncRNA,并通过顺式和反式作用预测其靶基因,对靶基因进行功能分析,构建关键lncRNA的共表达网络,分析并筛选毛囊周期性生长发育相关的关键靶向关系对。结果表明,lncRNA-MSTRG.13836.1表达量在脱毛组和不脱毛组具有显著差异,其顺式和反式靶基因与雌激素信号通路相关。构建了lncRNA-MSTRG.13836.1的共表达网络并筛选到靶向4个miRNAs及5个与毛囊周期性生长发育相关的基因,这些基因在Wnt、TGF-β、PI3K-Akt及MAPK等与毛囊周期性生长发育密切相关的信号通路中起调控作用。综上,lncRNA-MSTRG.13836.1对绵羊毛囊周期性生长发育具有调控作用,lncRNA以及靶向的4个miRNAs(miR-214-z、miR-214-y、miR-9298-x、miR-11977-z)和5个基因(PTCH2、CTNNB1、FZD3、Lpar6、Wnt10b)可作为绵羊毛囊发育相关的关键调控因子。 In order to analysis the function of lncRNA-MSTRG.13836.1 and to identify crucial target relationship pairs associated with cyclic growth and development of Dorper sheep hair follicle,eight 2-year-old female Dorper sheep were selected for this study,with 5 shedding sheep as test subjects and three non-shedding sheep as controls.Their skin samples were collected at 3 time points and subjected to RNA sequencing(RNA-seq).Crucial lncRNAs were screened by differential expression analysis,and their target genes were predicted by cis-role and trans-role,functional analysis of target genes was performed,co-expression network of crucial lncRNAs was constructed,and crucial target relationship pairs related to hair follicle cyclic growth and development were analyzed and screened.The results showed that lncRNA-MSTRG.13836.1 was significantly different between the hair shedding group and the non-shedding group,and its cis and trans target genes were associated with Estrogen signaling pathway.LncRNA-MSTRG.13836.1 co-expression network was constructed and screened to target 4 miRNAs and 5 genes related to hair follicle cyclic growth and development.The lncRNA-MSTRG.13836.1 was found to play a regulatory role in Wnt,TGF-β,PI3K-Akt,MAPK and other signaling pathways that were closely related to hair follicle cyclic growth and development.In summary,lncRNA-MSTRG.13836.1 had a regulatory role in sheep hair follicle cyclic growth and development,and the lncRNA,as well as 4 miRNAs(miR-214-z,miR-214-y,miR-9298-x,miR-11977-z)and 5 genes(PTCH2,CTNNB1,FZD3,Lpar6,Wnt10b)could be crucial regulators associated with hair follicle development in sheep.
