摘要
为探究类鼻疽伯克霍尔德菌噬菌体的生物学特性,本试验采用双层平板法分离筛选类鼻疽伯克霍尔德菌噬菌体;采用形态学、一步生长曲线、宿主范围测定分析该噬菌体的生物学特性;通过小鼠体内杀菌试验和体外消毒能力检测评估该噬菌体的疗效。结果显示,成功分离出1株类鼻疽伯克霍尔德菌噬菌体,命名为HNS1。形态学观察显示,噬菌体头部为正二十面体,直径为(62.8±3.0)nm,尾部长为(54.9±5.0)nm,根据命名法则该噬菌体属于肌尾噬菌体。一步生长曲线测定结果显示,该噬菌体的潜伏期约为32 min,释放量约为8.04 PFU/infection center。宿主范围测定结果显示,该噬菌体的裂解率达到73.3%(11/15)。小鼠体内杀菌试验和体外消毒能力检测结果显示,该噬菌体能够有效在小鼠体内和环境中裂解类鼻疽伯克霍尔德菌。结果表明,筛选得到的噬菌体具有较广的裂解谱,可能成为治疗类鼻疽伯克霍尔德菌感染的潜在抗菌剂。
To determine the biological characteristics of Burkholderia pseudomallei(B.pseudomallei)phage,this study used double-layer plate method to isolate and screen for phage,analyzed the biological characteristics of the phage by morphology,one-step growth curve,and host range,and evaluated the efficacy of the phage by in vivo bactericidal experiments in mice and disinfection tests in vitro.The results showed that one bacteriophage,named HNS1,was isolated from B.pseudomallei.Morphological observation showed that the phage had an icosahedral head with a diameter of(62.8±3.0)nm and a long,flexible tail with a length of(54.9±5.0)nm.According to naming conventions,this phage fell within the Myoviridae family.The one-step growth curve indicated that the latent period of the phage was about 32 min,and the burst size was about 8.04 PFU/infection center.The host range test showed that the phage had a lytic rate of 73.3%(11/15).In vivo bactericidal experiments in mice and disinfection tests in vitro demonstrated that the phage effectively lysed B.pseudomallei in mice and in the environment.Thus,the isolated phage has a broad lytic spectrum and offers a potential antibacterial agent for the treatment of B.pseudomallei infections.
