人参皂苷Rh2增强人肺腺癌顺铂耐药A549/DDP细胞顺铂敏感性的机制研究

摘要 目的探究人参皂苷Rh2提高A549/DDP细胞顺铂(cisplatin,DDP)敏感性的作用机制。方法MTT法检测DDP和人参皂苷Rh2对A549及A549/DDP细胞的抑制率和耐药指数;将A549/DDP细胞分为对照组、DDP组、人参皂苷Rh2组和人参皂苷Rh2联合DDP组,药物处理24 h后,流式细胞术检测细胞凋亡率;Western Blot法测定Bcl-2和Bax的表达水平。结果DDP组、人参皂苷Rh2组和人参皂苷Rh2联合DDP组均抑制了A549/DDP细胞的增殖,其中人参皂苷Rh2联合DDP组对A549/DDP细胞的凋亡率高于其他给药组。Bax蛋白表达水平明显提升,Bcl-2蛋白表达量显著降低。结论人参皂苷Rh2增强A549/DDP增殖对DDP的敏感性,诱导A549/DDP细胞凋亡的机制可能与干预Bax、Bcl-2蛋白表达有关。 Objective To investigate the mechanism of ginsenoside Rh2 on improvement the sensitivity of cisplatin(DDP)in human lung adenocarcinoma A549/DDP cells.Methods After 24 h treatment of DDP and ginsenoside Rh2,the inhibition rate and drug resistance index on A549 and A549/DDP cells were measured by MTT method.The A549/DDP cells were divided into control group,DDP group,ginsenoside Rh2 group and ginsenoside Rh2 combined with DDP group.The apoptosis was detected by flowcytometry.The expression levels of Bcl-2 and Bax were determined by Western Blot.Results The proliferation of A549/DDP cells was significantly inhibited in the DDP group,the ginsenoside Rh2 group and the ginsenoside Rh2 combined with DDP group,among which the apoptosis rate of A549/DDP cells was higher in the ginsenoside Rh2 combined with DDP groups than in the other administration groups.Conclusion Ginsenoside Rh2 enhances the sensitivity of DDP in A549/DDP,and the mechanism may be related to the protein expression of Bax and Bcl-2.
